Experimental Study On Apoptosis Effect Of Chinese Medicine Qingdushuan On SiHa Cells Of Human Cervical Cancer

Purpose: By the experimental research on the apoptosis which from traditional Chinese medicine Qingdushuan in vitro on human cervical cancer SiHa cells,To explore the mechanism of traditional Chinese medicine qingdushuan prevention and treatment of cervical cancer,to further improve the theoretical basis for the clinical application of traditional Chinese medicine Qing qingdushuan.Method:1.Extracting the effective ingredients of traditional Chinese herbal medicine Qingdushuan by chloroform extraction method.2.Human cervical cancer SiHa cells was cultured in vitro,Traditional Chinese medicine Qingdushuan assigned into dosage of high,middle and low,Acting upon the cervical cancer cell SiHa respectively at 24 h,48h,72 h,seting one blank control group and Baofukang suppository group,By CCK-8 assay to detect the effection on proliferation for high,middle and low groups of Qingdushuan acting on cervical cancer SiHa cells in vitro with different concentrations and different time points,Detect the optical density(OD),and determine the optimal time and effect point,to support the next experiment.3.Apoptosis of experimental groups were detected by mitochondrial membrane potential:The use of JC-1 mitochondrial membrane potential detection kit to detect the proliferation and inhibitory state after the different concentrations of Chinese medicine Qingdushuan effect on cervical cancer SiHa cell,thus further understanding of traditional Chinese medicine Qing dushuan induced cell apoptosis pathway.4.Apoptosis assay by flow cytometry assay: to detect the apoptosis effect after different concentration of Chinese medicine Qingdushuan effect on cervical cancer SiHa cells and detect the impact on the cell cycle by using fluorescent probe annexin-V/PI kit.Result: High dose of Chinese medicine Qingdushuan has obvious inhibition and inducing apoptosis of human cervical cancer SiHa cells in 48 h and Block the cell cycle in G0/ G1 phase,The blank control group compared with middle and low dose group of Chinese medichine Qingdushuan,have significant difference(P < 0.01),the high dose group of Qingdushuan compared with Baofukangshuan have no significant difference(P > 0.05).Conclusion: Chinese medicine Qingdushuan can promote the apoptosis of cervical cancer SiHa cell proliferation inhibition,so as to achieve the prevention and treatment of cervical cancer maybe by block the cell cycle in G0 / G1 phase.
