PURPOSES:To investigate the expression of ezrin and moesin in squamous carcinoma of cervix(SCC)and SiHa cells,and their correlation with invasion and metastasisl.METHODS:The expression of ezrin and moesin mRNA and protein were respectively detected by RT-PCR and immunohistochemistry in 20 cases of normal cervical epithelium(NCE),36 cases of cervical intraepithelial neoplasm(CIN),33 cases of SCC with opterative treatment(SCC-O)and 20 cases of SCC with radiative and/or chematherapy treatment before opteration(SCC-RCO).The reasonable concentration of cisplatin or taxinol to inhibit SiHa cell proliferation was assayed by MTT.The expression of ezrin and moesin mRNA were detected by RT-PCR after SiHa cells exposed to cisplatin or taxinol.RESULTS:1.The positive expression of ezrin and moesin gradually increased from NCE to CIN,and then to SCC-RCO and SCC-O(all P<0.05).In the cases with pelvic lymph node metastasis,deeper stroma involvement and intravasclar infiltration,the expression of ezrin was significantly higher than those in the cases without them(all P<0.05).In the cases with histolgical gradeⅢ,pelvic lymph node metastasis,deeper stroma involvement and intravasclar infiltration,the expression of moesin was significantly higher than those in the cases without them(all P<0.05).In the CIN and SCC groups,the positive staining reaction of ezrin and moesin were mainly located in the cell endochylema,but in cell membrane the staining reaction weakened or vanished. MTT assay showed that nearly 50 percent of SiHa cells was inhibited when they were cultivated after 72 hours,in 10mg/L DDP or in 15ug/L taxinol.The level of ezrin and moesin mRNA was significantly decreased with administering cisplatin or taxinol(P<0.05).2.The level of moesin mRNA was significantly lower than ezrin mRNA in the same group(P<0.05).The expression of ezrin in SCC-O was significantly positve related to moesin(r=0.8715,P<0.05).In the patients of ezrin and moesin positive,the prevalences of deeper stroma involvement and intravasclar infiltration were statistically higher than those in the patients with ezrin and moesin negative and with one of them being positive,respectively.(P<0.05).CONCLUSION:The over expression of ezrin and moesin is correlated with the squamous carcinoma of cervix development,invasion and metastasisl.Ezrin and moesin cytoplasmic mislocation were related to canceration.X-irradiation and chemotherapy could down-regulate the level of ezrin and moesin mRNA,which might suggest that ezrin and moesin take part in the course of cell apoptosis.
