There are about 56 species of genus Leontopodium(Compositae) all over the world and more than 41 grown in North China, Northwest and Southwest regions, especially in the Tibetan Plateau. About 25 species of Leontopodium are traditional Chinese medicine with anti-inflammatory activity for releasing cough, as expectorant and for the treatment of bronchitis, diarrhoea, dysentery, proteinuria and hematuria.We studied the chemical constituents of the roots of Leontopodium pusillum which was collected from Qinghai Province, 14 compounds have been isolated by means of silica gel column chromatography(CC), preparative thin layer chromatography(PTLC) and recrystallization. The structures of 10 compounds have been elucidated on the basis of spectroscopic methods(EMIS、IR、MS、1H-NMR、13C-NMR、2DNMR) and by comparison with authentic samples or with the data from the literatures, They are stigmasterol(1.1), 5-Metheoxyobliquin(1.2), Maniladiol palimate(1.3), Estigmast-4-en-6β-ol-3-ona(1.4), β-sitosterol(1.5), Alkyls caffeate(1.6),(3β)-3-Hydroxyolean-12-en-28-oic acid(1.7), Vitexilactone(1.8),((4aR,5S,9aR)-4a,5,6,7,8,9a-hexahydro-9a-hydroxy-3,4a,5-trimethyl-naphtha[2,3-b]furan-2(4H)-one)(1.9), Cryptomeridiol(1.10). Compounds 1.1 to 1.10 were first isolated from the Leontopodium pusillum, three sesquiterpenoids compounds were isolated from the plants of the Leontopodium for the first time. They are 1.8, 1.9, 1.10. In addition, their antimicrobial activity against Bacillus Subtilis, Staphylococcus Aureus and Escherichia Coli were tested, using the Oxford cup method. Antibacterial experiments showed that: the compounds 1.8-1.10 have no inhibitory effect on E. coli, while good inhibition on Bacillus subtilis and Staphylococcus aureus,especially 1.10, and the other testing compounds have no effect on the three strains.To search for new active ingredients, developt and utilize Leontopodium, we studied the chemical constituents of the essential oil of Leonfopodium calocephalum, which obtained by water vapor distillation and analyzed by GC-MS, of which 44 compounds have been identified. In addition, the essential oil antimicrobial activity against Staphylococcus Aureus and Escherichia Coli were tested, using the Filter method. Antibacterial experiments showed that: the essential oil of Leonfopodium calocephalum of experimental bacteria, there is no good antimicrobial activity.Results in this thesis are important for Leontopodium and chemtaxonomy, developing and utilizing the natural resources and screening new medicines.
