Trifolium pretense L. which belongs to Leguminosae sp. is an annual or perennial herb.There are many aliases such as clover,red clover,Holland and so on. Trifolium pretense L. is widely distributed in the world, second only to the same coverage area of Trifolium repens,which is the world famous cover plant. It is widely planted in Russia United States and other places. In China, Trifolium pretense L. is mainly distributed in Hubei, Xinjiang, Yunnan and other places, which is rich in resources. Chinese folk treat the inflorescence of Trifolium pretense L. as medicine, mainly used for cough, asthmatic and antispasmodic. And it is also made into an ointment, as local ulcer treatment available,which is blindly traditional folk medicine. In recent years isoflavones as the main chemical constituents of Trifolium pretense L. which its activity is deeply researched, found that it has a good estrogen-like effect, the role of anti-osteoporosis, anti-cancer role of biological activity.Objectives: Firstly to enrich the isoflavone of Trifolium pretense L. then study the pharmacological activity of the total extract enrichment. Finally Separate and purify the chemical components of the total extract with pharmacological activity.Methods: Trifolium pretense L. was extracted by 70% Et OH. The resulting total extract a macroporous resin with pure water and 70% Et OH elution, collection of 70% Et OH elution and concentration of 70% ethanol extract, and then extract polyamide column with water and70% Et OH elution, general provided material vacuum drying to obtain soil yellow powder.Study on the pharmacological activity of the obtained soil yellow powder in the treatment of osteoporosis. Separate and purify the chemical components from the site with pharmacological activity, according to the physical and chemical properties of LC-MS, IR,UV, 1H-NMR, 13C-NMR and other techniques for the structural characterization of the monomer compounds.Result: The pharmacological activities of the total extracts have good anti osteoporosis effect. Seventeen compounds had been isolated from the total extracts of Trifolium pr etense L. They are biochanin A(1)?formononetin(2)?irilone(3)?(+)-4-hydroxy-3-metho xy-8,9-methylene diooxypterocarpan(4)?maackiain(5)?biochanin-A7-O-?-D-glucopyrano side(6)?trifolirhizin(7)?ononin(8)??-sitosterol(9)?taxifolin(10)?quercitrin(11)?genist in(12)?Luteolin(13)?daidzein(14)?quercetin(15)?genistein(16)?daidzin(17);Through the analysis of the effective parts of the HPLC atlas research, there are 22 peaks in the HPLC atlas research, 16 peaks are obtained, and most of the effective parts are separ ated.Conclusion: Seventeen compounds had been isolated from the total extracts of Trifolium pretense L. Compounds 10 ? 13 are isolated from Trifolium pretense L. for the first time.Among the 17 compounds mostly are isoflavones. Through the analysis of the effective parts of the HPLC fingerprint, there are 22 peaks in the HPLC atlas research, 16 peaks are obtained, and most of the effective parts are separated.
