Atalantia buxifolia(Poir).Oliv.Alias:Atalantia,which is the underground root bark and rhizome of the plant Severinia buxifolia(Poir)Ten from the Rutaceae.It’s mainly distributed in Guangdong,Guangxi and Hainan province in China.It has the characteristic with pungent,bitter and slightly hot in the traditional Chinese medical theory. In many southern parts of China,the underground root of Atalantia buxifolia(Poir).Oliv. has been used to treat chronic bronchitis for many years with good effect. The modern pharmacological research has shown that it has strong effect to relieve cough and asthma and preventing phlegm from forming.Our lab has found that the n-Butylalcohol part of the 95% ethanol extraction had good activity to relieve cough and asthma.On the bases of that,we have studied the chemical constituents of effect parts and found the major are acridone alkaloids. This paper is aimed to figure out the chemical constituents of n-Butylalcohol effect part and the total alkaloids part to make clear the pharmacodynamic material basis of relieving cough and asthma activity.The main contents are as follows:1.Study on the constituents of n-Butylalcohol effective part :As the preview shows,the common Si O2 silicagel column adsorption chromatography, TLC chromatography, MCI chromatography,Sephadex LH-20,ODS-C18,preparative chromatography,recrystallization and preparative liquid chromatography all have been used to separate the chemical constituents.2. Study on the constituents of the Atalantia buxifolia total alkaloids. As the effective parts have shown that the main constituents are alkaloids,we used the same separationmethods to study the constituents of total alkaloids part.Research results:1. 12 compounds were separated and 9 compounds are identified from n-Butylalcohol part,and the identified compounds are as follows: Trans-N-p-coumaroyl tyramine(1);Artamenone(2);Citrusinine-II(7);Glycofolinine(8);1,3,5-Trihydroxy-4-(γ,γ-dimethylallyl)-a cridone(9);Cycloseverinolide(12);Auraptene(13); Severinolide(14);Dihydroisoartonin E-2’-methylether(15). The compounds(1) 2,(15) are the first time to be found in this plant.2. 8 compounds were separated and identified from the total alkaloids as follows:Buxifoliadine-B(3);Atalaphylline(4);N-methyl-Atalaphylline(5);Buxifoliadine-A(6);β-sitost erol(10);Stigmasterol(11);Ataphillinine(16);Glycocitrine-I(17).In this paper,the total alkaloids in effective parts were studied to lie a solid foundation for the further development of new drug research.
