ObjectiveTo research the effects of Trifolium Pretense Isoflavones (TPIF) on osteoporosis in mice(rats),and explore its possible mechanisms.Methods1. The prevention and treatment of TPIF on osteoposis in RA rats: sixty mice were chosen and were divided into six groups(control group,model group,vitamin D3 group and TPIF 50,100,200mg·kg-1 groups).Except model group administrated with distilled water,other groups administrated with RA.At the same time TPIF and Vitamin D3 were administrated. Serum biochemical indicators were measured and features of femoral bone were also observed.2. The prevention and treatment of TPIF on osteoposis in ovariectomized rats: sixty female Wistar rats were randomly assigned to six groups (n=10): sham operation group, osteoporotic model group, nilestriol 2.5mg·kg-1 group and TPIF 30,60,90mg·kg-1 groups. These experimental rats with the exception of sham operation group were ovariectomized and then administrated with TPIF or nilestriol for 3 months after 30 days of the ovariectomized operation. The rats were then killed and blood was taken for measurement of serum Ca2+, P3+, ALP, StrACP, BGP, E2. Body weight, BMD, bone biomechanics, pathomorphism of tibia were observed as well.Results1. The prevention and treatment of TPIF on osteoposis in RA rats: the weight of RA mice decreased,this may be related to the toxic effects of RA and drugs. The ALP, BGP and StrACP increased in OP mice.This is similar to the high turnover osteoporosis.The ALP, BGP and StrACP had been decreased significantly after TPIF administration(P<0.05 or P<0.01).Furthermore, E2 had increased obviously(P<0.05). Under the microscope,the bone cortical became thinner and the bone trabecula became sparser in OP mice.The bone trabecula were prevented obviously by administrating with TPIF.2. The prevention and treatment of TPIF on osteoposis in ovariectomized rats: the body weight induced by OVX was prevented obviously by administrating with TPIF. BMD had increased (P<0.05 or P<0.01). Furthermore, biomechanical characters of bone were measured by three point bending test, and the maximum loading and maximum stress increased remarkably(P<0.05). Serum E2 and BGP were examined by RIA, and results showed that E2 increased(P<0.01) and BGP decreased. The ALP, BGP , StrACP and urine Ca, P decreased obviously(P<0.05 or P<0.01). Under the microscope,the bone trabecular reduced significantly and bone marrow cavity filled with a large number of fat cells in OVX rats.However, the status reversed obviouslv by administrating with TPIF.Conclusion1. RA and ovariectomy induced osteoporosis were a high turnover osteoporosis and the two osteoporosis could reduce estrogen levels.2. TPIF could significantly improve the femur and tibia organization structure, increase bone mineral density and bone mass, enhance the biomechanical characters of femur.3. TPIF could significantly decrease the ALP, BGP and StrACP levels, reduce bone transformation, inhibit bone resorption and reduce bone loss.4. TPIF could prevent and treat osteoporosis. Its mechanism may be related to increase estrogen levels , enhance response of endogenous estrogen, inhibit bone resorption and promote bone formation.
