Objective:Study the relation between kin17 protein expression and clinical features and prognosis of patients with rectal cancer.Methods:1.Collected clinical features of 135 cases of rectal cancer patients who came from the first affiliated hospital of Nanchang university and were diagnosed the rectal cancer from November 2008 to November 2009.Collected the paraffin samples of 135 cases of rectal cancer patients,including tumor tissue and tissue adjacent to carcinoma.2.Using immunohistochemistry(IHC)to detect kin17 protein in rectal paraffin sample,and analysis the relation between kin17 protein expression and clinical parameters and prognosis.3.Using spss22 statistical software for analysis.chi-square test,fisher's exact test were used to analysis the relation between the expression of kin17 protein and clinical features.Kaplan Meier method,method of the Log – rank were used to analysis the single factor of 5 years of survival,multi-factor was analyzed by the COX regression analysis.Results:1.Kin17 protein expressed both in rectal cancer tissue and tissue adjacent to carcinoma.20 cases were highly expressed in tissue adjacent to carcinoma,76 cases were highly expressed in rectal cancer tissue.There was significant difference.2.The expression level of kin17 are statistically significant related with penetration depth(P<0.002),lymph node metastasis(P<0.0001)and distant metastasis(P<0.0001)? TNM(P<0.0001),pathological grade(p<0.046),vascular invasion(P<0.0001)and peripheral nerve invasion(P<0.006),There was no significant difference in age(P=0.602),gender(P=0.484),tumor location(P=1),and tumor style(p=0.068).3.univariate survival analysis showed that age(P = 0.015)?penetration depth(P<0.002),lymph node metastasis(P < 0.0001),distant metastasis(P < 0.0001)?TNM(P < 0.0001)and vascular invasion(P = 0.04),peripheral nerve invasion(P<0.003),CEA(P < 0.0001),CA199(P < 0.0001)tumor style(P = 0.02),.Multifactor analysis showed that the expression of kin17 protein?age ?CEA?lymph node metastasis?distant metastasis?tumor style are independent factors.Conclusion:1.Kin17 protein expression in rectal carcinoma tissue was significantly higher than normal tissue.2.The expression level of kin17 protein was positively correlated with penetration depth? lymph node metastasis?distant metastasis ? TNM?pathological grade?vascular invasion and peripheral nerve invasion.3.The high expression of Kin17 protein is the independent poor prognostic factor.
