Objective: The experimental study of rectal tissue through PTEN and MMP-7 expression and its relevance to explore the PTEN and MMP - 7 in the development of cancer of the possible role for cancer diagnosis and determine the biological properties of molecular biology detection indicators.Methods: From July 2007 to September 2008 surgical resection specimens of 40 cases of rectal cancer (adenocarcinoma of the rectum) and 30 cases of cancer near tissue were collected in Second Affiliated Hospital of Nanchang University ,of which 26 cases were male, 14 cases female, The age was 21 to 82 years old, average age was 54±1 years old; Histological grade: among which 17 cases were well-differentiated and poorly 23 cases were differentiated。25 cases without lymph node metastasis, 15 cases with lymph node metastasis, All of the patients did not accept the pre-operative radiotherapy and chemetherapy, the specimens were fixed by10 percent the conventional formaldehyde, paraffin-embedded, 4 um serial sections were cut , for HE staining and immunohistochemistry staining respectively,。The expression of PTEN and MMP-7 was detected by immunohistochemical method in all of the specimensResults :1 The expression of PTEN gene in Cancer tissues reduced to 57.5% (23/40), and MMP-7 expression was significantly increased to 75% (30/40).( P <0.05).2 PTEN in cancer and near cancer normal tissues were 20%(6/30) MMP-7 in cancer and nea rcancer normal tissues were 83.33%(25/30)3 The MMP-7 expression in Cancer has nothing to do with tumor size, tumor morphology in general, Lauren classification (P>.0.05)., The MMP-7 expression positive rate also increased (P <0. 05) With the increase in rectal cancer staging; In cancer with lymph node positive .The MMP-7 expression was significantly higher than that without lymph node metastasis (P <0. 01); The depth of local infiltration to the rectal wall, the higher expression of MMP-7-positive rate more. there is a significant difference (P <0. 01). the low of differentiation of the cancer cell positive rate is lower than that in the high and middle differention.The high there was a significant difference (P <0.01)4 The expression of PTEN protein mainly located in the nucleus, as a clear brown colour . There were nothing to do with tumor size, tumor morphology in general, Lauren classification in the abnormal expression of PTEN protein. ThePTEN protein expression In the low group of undifferentiated cancer intensity was significantly lower than in well-differentiated group (P <0. 05); ThePTEN protein expression with lymph node metastasis group was significantly lower than without lymph node metastasis group(P <0 . 05); With increase of the infiltration and the progress of clinical stages, PTEN protein expression significantly reducted in intensity (P <0.05) Along with the increase of stagese, the PTEN protein expression intensity also obviously decrease(P<0.05)5 PTEN and MMP-7 expression was negatively correlated (r =- 0.3796 p <0.05)Conclusion:1 Lauren classification and tumor size, differentiation were not related to PTEN and MMP-7 positive expression rate (P> 0.05); with tumor depth of invasion, lymph node metastasis and TNM stage (P <0.05).2 PTEN tumor suppressor genes in rectal cancer and during the development of the rectal cancer decrease or loss expression, and promote the development of cancer, metastasis and invasion. PTEN protein expression can be used as judying the biological behavior of colorectal cancer and determine the clinical parameter3 MMP-7 has a strong activity of matrix degradation.its increase in the expression shows the development of cancer, metastasis and infiltration4 PTEN and MMP-7 showed significantly negative correlation.
