Luliconazole is a new kind of antimycotic biosynthesis.A large number of experiments show that the antifungal activityof Luliconazole is better than usual antifungal drugs.At present, this drug ispreparedfor Luliconazole cream abroad and not listed in our country. Ethosome is a new transdermal drug delivery system, which is a multi-layer vesicle structure prepared by adding high concentration ethanol in liposome. It has better encapsulation efficiency and deformation. Compared with common liposome, the transdermal permeation rate was significantly increased, and more effective in delivery of drugs through the skin.So, it has become a hot spot in the study of transdermal drug delivery. We have successfully studyed the prescription ratio of Luliconazole ethosome,and the transdermal effect and antifungal activity of Luliconazole ethosome were evaluated in the laboratory.Now, we make a research for antifungal efficacy of Luliconazole ethosomes on animal model, which will lay a foundation for clinical trials.Purpose To establish an animal model of dermatophytosis and to evaluate antifungal efficacy of Luliconazole ethosomes on guinea pig tinea corporis model.Methods Animal models of dermatophytosis were established by inoculating dermatophyte suspension onto abraded skin on the back of guinea pigs.Thirty-two nealthy guinea pigs were randomly and equally divided into 4 groups, namely Luliconazole ethosomes group(A),Luliconazole liposome group(B),Luliconazole aqueous alcohol solution group(C) and control group(D). The therapeutic effect was evaluated according to skin lesion score and fungal examination results on the 7th,11th and 15 th day.Results Obvious lesions were observed and fungal examination was positive in control group on the 7th,11th and 15th day.The skin lesion score and mycological cure rates of Luliconazole ethosomes group on day 7,11 and 15 are 6.63±0.92 (0)?3.38±0.52 (87.5%)?0.88±0.64(100%).respectively.The skin lesion score and mycological cure rates of Luliconazole liposome group on day 7,11 and 15 are 7.00±0.53 (0)?4.88 ± 0.35 (75%)?1.00±0.76 (100%), respectively. The skin lesion score and mycological cure rates of Luliconazole aqueous alcohol solution group on day 7,11 and 15 are 6.75 ±0.71 (0)?4.68±0.64 (75%)?1.00±0.53 (87.5%), respectively.The skin lesion score and mycological cure rates of control group on day 7,11 and 15 are 6.68±0.64(0)?6.93 ±0.52 (0)?7.13±0.64 (0) respectively.On the 11 th day,there was significant difference in clinical and mycological efficacy between the A, B, C group and D group (all P<0.05).And the clinical efficacy of A group is better than Bgroup and C group(all P<0.01).There was no significant difference in mycological efficacy amongA,B and C group(P>0.05).On the 15th day, there was significant difference in clinical and mycological efficacy between the A, B, C group and D group (all.P?0.05).Conclusion Luliconazole ethosomes,Luliconazole liposome and Luliconazole aqueous alcohol solution exhibit excellent antifungal activity in murine model of T.mentagrophytesand.The antifungal activity of Luliconazole ethosomes is better than Luliconazole liposome and Luliconazole aqueous alcohol solution.
