Objective: Alzheimer's disease(Alzheimer's disease,AD), is a kind of diseases onset in old age which is primary degenerative brain disease, mostly hidden onset, progressive cognitive dysfunction and abnormal spirit as the main clinical manifestations.The exact etiology and pathogenesis of AD is unknown, But persistent inflammatory reaction is one of the important mechanisms for the formation and development of AD. This subject adopts the bilateral hippocampal microinjectionin within A?1-42 to the preparation of AD model rats, by observing the electroacupuncture for AD model rats behavior change, the level of inflammatory factors serum and brain, To explore the role of astrocytes in AD model rats and the effect of acupuncture intervention, in order to provide a new basis for the treatment of AD.Methods: 40 healthy male Wistar rats, were randomly divided into normal group, electroacupuncture group, Sham operation group, model group. 10 rats in each group. AD rat model was prepared by micro-injection of A?1-42 into rat hippocampus, and the Sham operation group with normal saline instead of A ? 1-42 microinjection. The normal group, model group, the sham operation group without any treatment; The rats in the electroacupuncture group acupuncture "Baihui" and "Shenshu" point, USES the continuous wave, frequency of 5 Hz, the intensity of 1 m A, for 20 min, once a day, 6 days a course of treatment, a total of 2 courses of treatment, between 2 courses has a day to rest. After the electroacupuncture treatment,(1) using Morris Water Maze to observe the learning and memory ability of rats,(2) using the method of ELISA to detect the levels of serum Inflammatory factors IL-10? IFN-?,(3) The expression changes of IL-10 and IFN-?in rat hippocampus were detected by immunohistochemical method and Western-Blot method,(4) Using double immunofluorescence labeling method detection of rat hippocampal astrocytes GFAP + / IL-10 + expression changes.Results:(1) the behavioral observation: compared with model group, the electroacupuncture group escape latency was significantly shortened(P<0.01) the number of across platform obviously increased(P<0.05).(2) Serum inflammatory factors: compared with normal group, the model group serum IL- 10 decreased significantly(P<0.01), IFN-? increased significantly(P<0.05); Compared with model group, the serum levels of IL-10 electroacupuncture group was obviously increased(P<0.01), IFN- ? significantly decreased(P<0.05);(3) hippocampus inflammatory factors : compared with normal group, model group IFN- ? protein expression was significantly increased(P<0.05), IL-10 protein expression decreased significantly(P<0.01); Compared with model group, the curative group of IL-10 protein expression was significantly increased(P<0.01), IFN- ? protein expression decreased significantly(P<0.05).(4) Co-location expression of astrocytes and inflammatory factors: compared with normal group, model group GFAP+/IL-10+ expression increased significantly(P<0.05), compared with model group, the electroacupuncture group GFAP+/IL-10+ expression significantly increased(P<0.05).Conclusion: AD model rats pro-inflammatory factor IFN- ? expression increased, anti-inflammatory factor IL- 10 express decreased, with the excessive activation of astrocytes at the same time; Electroacupuncture can effectively improve the learning and memory ability of AD rats, increase the expression of anti-inflammatory factors IL-10, reduce the expression of pro-inflammatory factor IFN-?, and inflammatory factor levels in the brain and serum(central and peripheral) consistent trends; At the same time, activated astrocytes express anti-inflammatory cytokine IL-10. Indicated that Astrocyte activation is involved in the occurrence and development of AD, mediated by imbalance of pro-inflammatory/ anti-inflammatory factor; Electroacupuncture can down regulate the expression of proinflammatory cytokines and expression of anti-inflammatory cytokines, and activate the expression of astrocytes express anti-inflammatory factor, thus effectively inhibit AD inflammatory cascade development, To achieve the purpose of AD symptomatic improve.
