Objective:With the aging of the population, the incidence of Parkinson's disease (PD) is also rising. Clinical manifestations of PD include stilly shacking, bradykinesia, rigidity muscles and abnormal posture and pace. PD's mainly pathology change maintains the death of most of the dopamine neurons which from tight zone of substantial nigra of midbrain to striatum. Because the pathogenesis of PD is not clear, there is great difficulty in treatment, still no cure for Parkinson's disease. Dopamine replacement therapy is the current treatment of PD, replacement therapy only can relieve the symptoms but not to terminate or reverse the pathological process of PD, long-term use will inevitably be the emergence of drug resistance phenomenon and can cause serious side effects, so that sicker or create new symptoms, form a vicious cycle. In recent years, acupuncture as a unique, safe and convenient treatment method has been increasingly used in clinical, made a more positive effect. The experiment study the rats'rotation and the quantity of TH,Microglia,TNF-α,IL-1βand IFN-γin substantia nigra in order to study the therapeutically effect and mechanism of the electroacupuncture EA on Parkinson's disease(PD) which will offer experimental basis of applying acupuncture in clinic practice of the acupuncture.Methods:The Parkinsonian rats were modeled by stereotaxieal microquantitative injection in the right striatum with 6-hydroxydopamine and treated by the electroacupuncture therapy of Fengfu and Taichong point. All the experimental rats were divided into four groups:Normal Group, Sham-modeling Group, Model Group, Electroacupuncture Group.(1) Observation on behaviors:apomorphine was injected into abdominal cavity to induce left rotation. The number of circles rotated within 30 min after injection of apomorphine was recorded.(2) The therapy of immunohistochemistry is adopted to to observe the morphologic changes of substantia nigra and quantify the tyrosine hydroxylase (TH) and ionized calcium bindingadaptor molecule-1(Iba-1) positive neurons by the immunohistochemistry.(3) The therapy of immunohistochemistry is adopted to quantify TNF-α,IL-1β,IFN-γpositive cells in substantia nigra.Result:(1) In EA group, the number of rotations after the treatment was lower than before the treatment(P<0.05), but that didn't change in PD group(P>0.05). (2) The number of TH in PD group was obviously lower than that of the normal group(P<0.01), and that in the EA group was higher than the PD group(P<0.01),the number of microglia in PD and EA group was obviously higher than that of the normal and Sham-modeling group(P<0.05), the number of microglia in the EA group was obviously lower than of the PD group(P<0.05). (3) the number of TNF-α,IL-1β,IFN-γpositive neurons In PD group was obviously more than those of the normal group and sham-operation group (P<0.01).In the EA group, the number of TNF-α,IL-1β,IFN-γpositive neurons were less than that of the PD group(P<0.01).Conclusion:Neuritis response is an important pathogenesis of PD, Microglia cells in the nervous system plays an important role in the immune inflammatory. There is inflammation in model of Parkinsonian rats with 6-OHDA-mediated. Electro-acupuncture can significantly improve the rotational behavior of PD rats, Increased levels of TH in the substantia nigra, reducing the inflammatory response within the substantia nigra.
