Objective Poplar flower is dry trivia of salicaceae poplar plants,Canada poplar or other poplar plants,which can be used for curing diarrhea.However,to the best of our knowledge,the antioxidant and antimicrobial effects of Poplar flower flavonoids has not been demonstrated.Considering poplar flower as the research object,we aim to study on the antioxidant and antimicrobial effects of Poplar Flower flavonoids extracttion.Methods The ultrasonic extraction method to extract the flavone in the flower of poplar and the ultraviolet spectrophotometer method to test the flavonoids content were determined.Yield poplar flower flavonoids compounds as examining index,through the test to determine the most suitable solvent for poplar flower flavonoids compounds extraction,which is ethanol.Determined by single factor experiment,poplar flower flavonoids could be extracted by the best conditions of various factors.On the basis of single factor experiment,selected four factors influencing the most significant indicators,designed orthogonal test and filtrated the optimizaing ultrasonic extraction conditions of the poplar flower flavonoids.Extracted three different kinds of poplar flower compounds,through the drug susceptibility test we studied on escherichia coli,bacillus subtilis,staphylococcus aureus and salmonella bacteriostatic ring size,and used double dilution method to determine the minimal inhibitory concentration(MIC).By the methods of DPPH,the pyrogallol autoxidation method and methyl violet-Fe2 +-H2O2 reaction system combined spectrophotometer method,the antioxidant activity of poplar flower flavonoids were determined.Results Single factor experimental results showed that: the optimal ratio between material and liquid was about 1:20 ~ 1:30,optimal ethanol concentration was among 50% ~ 70%,the best extraction time was 90 ~ 150 min,the optimum extraction temperature was 50 ~ 70?.Chose the four factors of the ethanol concentration,solid-liquid ratio,extraction time and extraction temperature to design an orthogonal experiment.Through the orthogonal experiment optimizal extaction conditions,we got the optimal combination of temperature 70 ?,70%ethanol concentration,solid-liquid ratio 1:20 and the extracting time of 150 min.Flavonoid yield was 2.6% on average.The ultrasonic extraction was extremely sensitive to escherichia coli,was highly sensitive to salmonella and staphylococcus aureus and was mediumly sensitive to bacillus subtilis.Bacteriostatic effects of liquor extracts and alcohol extracts on escherichia coli was better than e.coli.The bacteriostatic effects of the ultrasonic extracts on those two kinds of bacteria was both the same.Bacteriostatic effect of ultrasonic extract on salmonella was better than water extract and alcohol extract.The poplar flower ultrasonic extraction could clearly eliminate DPPH free radical,·OH and ·O2-,which presented a certain dose effect.It showed better antioxidant activity than VE.While compared with VC,its clearance ability was weaker.Its antioxidant capacity was higher than the water extraction and the alcohol extraction.Conclusions The optimal extraction conditions for total flower of poplar extractions: temperature 70?,70% ethanol concentration,material-liquid ratio 1:20 and the extracting time 150 min.Flavonoid yield was 2.6% on average.The flower of poplar extract performed certainly antibacterial function on e.coli,bacillus subtilis,staphylococcus aureus and salmonella.The effects of e.coli was better than salmonella and staphylococcus aureus.The ultrasonic extracts showed weaker antibacterial function on bacillus subtilis.Antibacterial effect of the ultrasonic extraction was stronger than the water extract and the alcohol extract.All extraction samples performed great antioxidant activity,and in all kinds of extraction methods,the ultrasonic extract showed the most obviously antioxidant capacity.
