Coreopsis tinctoria flower(CTF)grown on Kunlun Mountain at an altitude of3000metres above sealevel. They have an effect on blood lipids and blood pressure. At present, lack of researches on differecesof physiological and biochemical due to ingredient differences and effects on the cellular and molecularlevel. In this study (Animal experiment), we are going to research the effect of aqueous and ethanolextracts of CTF on physiological and biochemical indexes of rats to compare the different from twodifferent methodologies. To study the effect of aqueous and ethanol extracts of CTF on the proliferationand differentiation of adipocytes in vitro, in order to provide based on cell biology of lowering mechanismby CTF.In Vivo experiment: The effect of aqueous extracts of CTF in high dose (CTF-AH) and low dose(CTF-AL), and the ethanol extract in high dose (CTF-EH) and low dose (CTF-EL) was given to SD ratsand body weight was weighed weekly. At day28after the blood pressure measuring all rats were sacrificedthe blood physiological parameters were tested. Results: the body weight of CTF-EH and CTF-EL rats wassignificantly decreased in comparison with that in CON rats (P<0.05), CTF-AH and CTF-AL rats weightchanged irregular; The blood pressure was not influenced by CTF treatment. Effect on bloodroutine, intermediate cell ratio in addition to KunLun snow chrysanthemum water extraction and low dosegroup, other groups were significantly lower than those in the control group; Compared with CON group,GLU content in three CTF groups (CTF-AL, CTF-EL, CTF-EH) was lowered by26%、38%、53%(P<0.01) and CHOL content increased extremely (P<0.01) at CTF-AL; it showed the decrease36%、37%ofTG at CTF-AL、CTF-EL (P<0.05); CTF-EL group was significant increase of HDL-C (P<0.05), othergroups had no significant difference. The two kinds of extracts of CTF treatment had no effect on LDL-C.In vitro experiment: Intervention primary preadipocytes by aqueous extracts and ethanol extractwith different doses. The proliferation of preadipocytes was detected bt MTT method, calculated the rate ofdifferentiation of cells in effects unit by observing, detect the content of intracellular and extracellular TG、LEP by ElISA Kit. Results: the extremely promoting effects of CFA and CFE were the best in30-50μg/mL group (P<0.01), the inhibit promoting in CFE-100μg/mL, there were no difference in other CFEgroups. CFA and CFE in30-100μg/mL have extremely Inhibit differentiation of preadipocytes (P<0.01);CFA in30-100ug/mL extracellular reduced the concentration of TG intracellular (P<0.05),TG ofextremely reduced only in CFA50-100μg/mL (P<0.01); LEP of extracellular was extremely reduced inCFE30μg/mL-100μg/mL (P<0.01).Conclusion:(1) CTF has no influenced on rat’s blood pressure and Effect on bloodroutine indexes were in the normal range,so as the health products are safe to use.(2) The aqueous extractsand ethanol extract of CTF inhibit the differentiation of preadipocytes, reduce the content of TG and GLUof serum,also reduce the content of TG and LEP of intracellular and extracellular, indicating that CTF mayhave a certain effect on the decrease of blood lipid in rats.
