Flavonoids are the compound in nature, with a structure of phenyl chromogen flavone. Their molecules have a keto form carbonyl group, and the first oxygen atom is alkaline, which is combined with strong acid to form salt. Hydroxy derivatives are also called yellow base element or yellow ketone, because they are yellow. Flavonoids are usually combined with sugar form flavonoid glycosides in the plant, a small flavonoids are in free form, and are mainly the O-glycoside and a little C-glycoside. Most of the plants contained flavonoids, which play an important role in plant growth, such as bacterial diseases and other aspects.Turmeric is a perennial herbaceous plant, widely distributed in many provinces and cities in our country. Turmeric has the reputation of "medicinal turmeric" and the very good development potential. It is reported in many thesises, the dioscin in turmeric is widely used in the preparation of some medicine, such as Anti-inflammatory, analgesic, cerebrovascular, birth control pills and so on. It is not reported turmeric also contains flavone glycosides, tannins, pigments besides saponin.Flavonoids with a variety of biological activity have very important significance in many diseases. Studying flavonoids has the vital significance in the turmeric. The flavonoids in the turmeric are studied systematically in this article.(1) In this paper, based on the Flickā€™s first law and extraction mechanism of Chinese herbal medicine, it is established dynamic model of the ultrasonic extraction of total flavonoids in the concentration of flavonoids in turmeric to get the relationship between the concentration of flavonoids and the ultrasonic time, ultrasonic power, considering the effect of ultrasonic field on solid liquid diffusion and ultrasonic wall-breaking effects on extraction yield of total flavonoids in turmeric. Then the total flavonoids in turmeric are extracted by the ultrasonic auxiliary. After numerical simulation of experimental data by Matlab software, the ultrasonic dynamic model of total flavonoids extracted from turmeric is verified.(2) There are some discussions about ultrasonic assisted extraction process optimization of total flavonoids in turmeric by orthogonal design of experiment. On the premise of single factor experiment, orthogonal experiment of four factors three levels is designed and analyzed comprehensively, selecting different ultrasonic power, solid-liquid ratio, ultrasonic time and different extracting liquid concentration. And the effect of the factors to extraction flavonoids is solid-liquid ratio>ultrasonic power>ethanol concentration>ultrasonic time. The best technological conditions of extracting total flavonoids in turmeric is ultrasonic power480w, solid-liquid ratio1:50ml/g, ultrasonic extraction time30min, ethanol concentration80%. In this condition, after the three parallel experiments, the average of extraction efficiency of flavonoids in turmeric is11.18mg/g.(3) Using Box-Behnken Design-response surface optimization method, based on the single factor experiment, ultrasonic power, liquid-solid ratio, ultrasonic time and ethanol concentration are selected to design Box-Behnken Design experiment. And by the four factors three levels of response surface analysis method the effect of the factors to extraction is liquid-solid ratio>ultrasonic power> ultrasonic time> ethanol concentration. The best technological conditions of extracting total flavonoids in turmeric are ultrasonic power400.76w, liquid-solid ratio1:42.19g/ml, ultrasonic time29.53min and ethanol concentration79.60%. Theoretically extraction yield of total flavonoids is11.23mg/g. The optimization results is verified,considering the practical operational problem, saving time, the energy consumption and achieving the best effect. The best correctional technological conditions of extracting total flavonoids in turmeric are ultrasonic power380w, liquid-solid ratio45:1g/ml, ultrasonic time30min and ethanol concentration80%. In the three experiments, the average of extraction efficiency of flavonoids in turmeric is11.59mg/g. They are no significant difference.So based on the response surface analysis the parameters of optimized extraction process is accurate, reliable and practical, which provides a theoretical basis for industrial mass production.
