Purpose: Impacts on the Contents of INF-? and IL-4 of IgA Nephropathy Model Rats Treated with the Therapy for Tonifying the kidney Bushenjianpi and JieduliyanMethod: A total of 80 healthy male Wistar rats of clean grade,weight(20020)g,one weeks after adaptive feeding,Using stratified random method rats were divided into 10 groups,model group 70.The model for bovine serum albumin(BSA)oral gavage,lipopolysaccharide(LPS)intravenous injection and carbon tetrachloride(CCL4)by subcutaneous injection of established IgA nephropathy rat model.Success after the model,The model group rats were redivided into model group,control group,Tripterygium Glycosides(TWP)group,Bushenjianpi and Jieduliyan high,medium and low dose group.Given the TWP control group was gavaged with TWP 6mg/(kg.d);Given the Bushenjianpi and Jieduliyan low dose group was gavaged with 4ml/(kg.d),Containing crude drug 8g/kg,the middle dose group was gavaged with 4ml/(kg.d),Containing crude drug 16g/kg,the high dose group was gavaged with 4ml/(kg.d),Containing crude drug 32g/kg.The control group and the model group was given saline gavage daily,4ml/rat Total of 8 weeks of administration.The observation index:(1)The general state of rats;(2)To test the 24-hour urine red blood cell count and 24-hour urine proteinuria quantitative changes;(3)Tested kidney function,liver function;(4)Were measured with ELISA method for the determination of serum and renal tissue IFN-?,IL-4;(5)Immunohistochemistry was used to observe the expression of IL-4,IFN-?and positioned in the renal tissue;(6)Using immunofluorescence to detect IgA complex deposition in the kidney tissue.Result:(1)During the experiment rats were killed 22,including 10 killed validate the model,modeling died during the 8,4 died during treatment(TWP control group died 3,Chinese medicine low dose group died 1).(2)Better blank group rats during the whole experiment,no obvious abnormality.The model group rats were given subcutaneous injection of carbon tetrachloride showed varying degrees of depression,hair messy and not shiny,mouth nose secretions,diarrhea,or reduce the slow increase of body weight.After 2~3 weeks after treatment,the general state of rats in each treatment group improved,Relieve diarrhea,weight gain.(3)The normal group rats without hematuria and proteinuria positive performance,The model rats 24-hour urine red blood cell count and 24-hour urinary protein excretion was significantly higher in each treatment group were decreased(P <0.05),no significant differences between the treatment group and the control group TWP between groups(P>0.05).(4)IgA nephropathy in renal tissue of rats in the control group and the level of IFN-? is relatively decreased,the level of IL-4 were significantly increased.(5)Each treatment group after treatment of rat IFN-? levels were significantly up-regulated(P <0.05),the level of IL-4 was significantly lower(P <0.05),Comparison between groups,Bushenjianpi and Jieduliyan high,medium and low dose group There was no significant difference between the control group and TWP(P> 0.05).(6)There was no significant difference between groups of renal function and liver function in rats(P> 0.05),but the TWP group had 2 cases of significant liver injury event.Conclusion:(1)Methods using bovine serum albumin gavage and carbon tetrachloride,LPS injection could successfully establish animal model of IgA nephropathy.(2)TWP group and Bushenjianpi and Jieduliyan group effectively reduce IgA nephropathy model rats 24 hours urine red blood cell count and 24 hours urinary protein excretion rate.(3)After testing compared with the control group,decreased the content of IFN-? in renal tissue of IgA nephropathy model rats,the IFN-? was significantly increased after drug treatment was positively correlated with the disease;renal tissue of IgA nephropathy rats increased after drug treatment,the decrease of IL-4,and negatively correlated with the disease;that the abnormal expression of IFN-?,IL-4 involved in the pathogenesis of IgA nephropathy.(4)Each treatment group,Bushenjianpi and Jieduliyan prescription possible mechanism by adjusting the level of Th type cytokines IFN-? and IL-4 in renal tissue of rats IgAN model,to delay the development of IgAN.(5)By comparison with the control group of rats rat liver and kidney function modules made no difference,but TWP group 2 patients significant liver damage event occurs.(6)Bushenjianpi and Jieduliyan law in reducing IgA nephropathy with hematuria,proteinuria,and TWP no significant difference in the control group,but no liver and kidney damage and other side effects,worthy of further research and extension.
