Objectives: To observe the effect of Yuye Decoction and its disassembled prescriptions on the related indexes of type 2 diabetic rats, such as weight, blood glucose, blood lipids, insulin levels, and inflammatory cytokines; To explore the mechanism of Yuye Decoction and its disassembled prescriptions on treating type 2 diabetes, and the compatibility of Yuye Decoction; To provide the oretical and experimental basis for using Yuye Decoction to treat diabetes.Method: Continuous high fat diet for 4 weeks, then replicate the type 2 diabetes mellitus model in SD rats through a single intraperitoneal injection which need 35 milligrams streptozotocin(STZ) per kilogram of weight. Type 2 diabetic rats were randomly assigned to the model group, Yuye Decoction group, The No.1 disassembled prescriptions group, The No.2 disassembled prescriptions group, Metformin group, and the blank control group. Drugs were given by intragastric administration to each group, once each day for 8 weeks. Then measure the weight and the blood glucose of each rat, And strictly in accordance with the relevant provisions of the kit to detect the levels of a full set of blood lipids(including TC, TG, HDL-C, LDL-C), inflammatory cytokines(including IL-1 beta, IL- 6, TNF- alpha) and insulin.Results: 1. Better general condition were observed in the Yuye Decoction group, the No.1 disassembled prescriptions group and the No.2 disassembled prescriptions group. After 8 weeks of treatment, all of the rats in the Yuye Decoction group, the No.1 disassembled prescriptions group and the No.2 disassembled prescriptions group, had better mental state than before. And there symptom of polyuria and polydipsia were improved. Colours of the tongue of rats in Yuye Decoction group and the No.2 disassembled prescriptions group turned into light red, while colours of the tongue of rats in the No.1 disassembled prescriptions group turned into slightly dark red, and the tongue of rats in Metformin group were still dark purple. After treatment, compared with model group, the treatment group of diabetic rats increased in weight which Yuye Decoction group and metformin group has very significant difference(P<0.01), No.2 disassembled prescriptions group has significant difference(P<0.05). The weight difference between the Yuye Decoction group and the metformin group was not statistically significant.2. All of Yuye Decoction, No.1 disassembled prescriptions and No.2 disassembled prescriptions had obvious hypoglycemic effect on type 2 diabetic rats.When the model of type 2 diabetic rats were successfully produced, compared with the normal group, the fasting blood glucose levels of the other rats were significantly increased(P<0.01). After 8 weeks of treatment, compared with the model group, the fasting blood glucose levels of each treatment group were significantly reduced(P<0.01). The fasting blood glucose of rats in Yuye Decoction group and the two disassembled prescriptions groups were higher than the the fasting blood glucose of rats in metformin group, but only the difference between the No.1 disassembled prescriptions group and the metformin group was statistically significant. The difference between Yuye Decoction group and the No.1 disassembled prescriptions group was also statistically significant.3. The abnormal lipid metabolism of type 2 diabetic rats in Yuye Decoction group and the two disassembled prescriptions groups were improved. After 8 weeks of treatment, compared with the model group, the total cholesterol and the triglycerides and the low density lipoprotein-cholesterol of each treatment group were decreased, the high density lipoprotein-cholesterol of each treatment group were increased. The overall effect of Yuye Decoction on regulating abnormal lipid metabolism is best, which is better than the disassembled prescriptions and the metformin,and the overall effect of the disassembled prescriptions on regulating abnormal lipid metabolism is better than the metformin.4. The levels of insulin in type 2 diabetic rats of Yuye Decoction group and the two disassembled prescriptions groups were significantly increased.After 8 weeks treatment, compared with the model group, the serum insulin levels of each treatment group were significantly increased(P<0.01); The Yuye Decoction group and the metformin group have the most remarkable effect on increasing the serum insulin levels, the effect of the metformin on increasing the serum insulin levels is superior to the effect of the two disassembled prescriptions.5. Yuye Decoction and the two disassembled prescriptions can significantly reduce the level of inflammatory factors in rats with type 2 diabetes.After 8 weeks of treatment, compared with the model group, the levels of Interleukin-1?, Interleukin- 6 and Tumor Necrosis Factor-alpha in rats of Yuye Decoction group and the two disassembled prescriptions groups were very significantly decreased(P<0.01), while only the level of Tumor Necrosis Factor-alpha were very significantly decreased(P<0.01) In the metformin group.Conclusion: Yuye Decoction and the two disassembled prescriptions can significantly improve the type 2 diabetic rats' blood glucose, blood lipid and inflammatory factor, and can enhance the serum insulin levels in type 2 diabetic rats. Although the two disassembled prescriptions were not as good as metformin in reducing fasting blood glucose and improving the insulin levels, but Yuye Decoction and the two disassembled prescriptions are superior to metformin in the regulation of lipid metabolism abnormality and reducing the level of inflammatory factors. There are a lot of differences between the effect of Yuye Decoction on general situation and objective index of type 2 diabetic rats and the effect of the two disassembled prescriptions on general situation and objective index of type 2 diabetic rats. This shows that, the prescription compatibility law of traditional Chinese Medicine plays an important role in the process of using traditional Chinese medicine in the prevention and treatment of diabetes.
