Study On Composing Mechanism By Effect Of Disassembled Compositions Of Yiqi Huoxue Decoction On Platelet Aggregation Of Health Adults

OBJECTIVE: To investigate and explain the composing mechanism by observing enhancement property of disassembled compositions of Yiqi Huoxue decoction (YQHXD) on platelet aggregation of health adults in vitro.METHODS: YQHXD was disassembled to 11 groups such as the whole prescription,the whole prescription without Glycyrrhiza uralensis Fisch,Astragalus membranaceus(Fisch.) Bge and Angelica Scinesis (Oliv.) Diels,Yiqi group,Huoxue group and every single herb. 72 of healthy human subjects were divided into 12 groups with 6 of each group at random. Venous whole blood was obtained from them and anti-coagulated with 1:9 Natrium Citricum, and then centrifuged to produce platelet-rich plasma (PRP) and platelet-poor plasma (PPP), and diluted the platelet concentration of PRP to 150×10~9/ L with PPP. Disassembled compositions of YQHXD and Sodium Chloride were placed in different translucent tubes containing PRP and PPP, calculated the maximum percents of platelet aggregation stimulated by platelet agonists: Adenosine Diphosphate (ADP), Platelet Activating Factor (PAF) and Arachidonic Acid (AA).RESULTS: Compared with the blank group, the maximum percents of platelet aggregation stimulated by ADP were largely improved by the whole prescription,the whole prescription without Glycyrrhiza uralensis Fisch.,Astragalus membranaceus(Fisch.)Bge and Angelica Scinesis (Oliv.) Diels,Huoxue group have significance in statistics(P<0.01),which haven't significance in statistics in Yiqi group and every single herb(P>0.05). The maximum percents of platelet aggregation stimulated by PAF were largely improved by the whole prescription,the whole prescription without Glycyrrhiza uralensis Fisch.,Astragalus membranaceus(Fisch.)Bge and Angelica Scinesis (Oliv.) Diels have significance in statistics(P<0.01, P<0.05), which were prominently declined by Yiqi group,Huoxue group,Codonopsis pilosula(Franch.) Nannf.,Prunus persica(L.) Batsch and Glycyrrhiza uralensis Fisch(P<0.01). The maximum percents of platelet aggregation stimulated by AA were dramatically depressed by Prunus persica(L.) Batsch(P<0.05) , which haven't significance in statistics in other groups(P>0.05).CONCLUSION: The maximum percents of platelet aggregation induced by ADP and PAF were largely improved by YQHXD, Both Astragalus membranaceus and Angelica Scinesis played the most important roles in the decoction, the herbs of Yiqi and Huoxue must work together to enhance platelet aggregation. In vitro study, YQHXD can markedly improve platelet aggregation of health adults, but the exact mechanism should be proved by further experiments.
