Experimental Study On The Effect Of Yuye Decoctionon On The Podocytes And Its Related Protein Expression In Rats With Type 2 Diabetic Nephropathy

Objectives:To observe the effect of Yuye Decoction(YYD)on 24 hours urinary albumin excretion rate,renal index,biochemical indicators(such as blood glucose,blood urea nitrogen,serum creatinine,etc.),the protein expression of nephrin and podocin,and the morphological changes of renal podocytes by electron microscope in rats with diabetic nephropathy,we evaluated the protective effect of YYD on renal podocytes in rats with diabetic nephropathy,and then explored its mechanism so as to providing some theoretical and experimental basis for clinic use.Methods:Diabetic nephropathy(DN)model was established by SD malerats with high-fat diet accompanied with intraperitoneal injection of streptozotocin(STZ)in the left lower abdomen.All rats were randomly divided into six groups,namely the control group,the model group,the low dose group of YYD(YYD-L),the medium dose group of YYD(YYD-M),the high dose group of YYD(YYD-H),the metformin group,that each one was made up of 8 rats.The rats in low,medium and high dose groups of YYD were administrated by gastric perfusion with the concentration of YYD of 2g/3ml,4g/3ml and 5g/3ml respectively.The rats of metformin group were administrated by gastric perfusion with the concentration of metformin of 90mg/3ml,and the rats in control and model group were administrated with3 ml distilled water,once a day for 8 weeks.At the end of experiment,the levels of 24 hours urinary albumin excretion rate,the renal index,blood glucose,blood urea nitrogen and serum creatinine were measured,the protein expressions of both nephrin and podocin in renal podocytes were detected by immunofluorescence semi-quantitation method,and the morphological changes of podocyte were observed by electron microscope.Results:1.24 hours urinary albumin excretion rate: The 24 hours urinary albumin excretion rate of the model group was obviously higher than that of the control group(P<0.05).The data of YYD-H group and metformin group were clearly lower than the model group(P<0.05).The data of YYD-L group and YYD-M group were lower than the model group.2.Renal index: The renal index of the model group wasobviously higher than that of the control group(P<0.05).The data of YYD-H group and the metformin group were clearly lower than the model group(P<0.05).The renal index of YYD-L group and YYD-M group were lower than that of the model group.3.Some biochemical indicators : Compared with the control group,blood glucose,serum creatinine and blood urea nitrogen were increased in the model group(P<0.05).The data of YYD-H group and the metformin group were lower obviously than the model rats(P<0.05).4.The pathological changes of renal tissue in rats by light microscope: The renal tissue structure of rats in the control group was clear and intact.The glomerular hypertrophy and tubular deformation were observed in the model group.Compared with the model group,the pathological changes of renal tissue were significantly alleviated in YYD-H group and the metformin group.5.The ultrastructure changes of podocytes under electron microscope: There were no distinct abnormal pathological changes in the control group.The typical pathological changes,such as fusion of podocytes,the decreasing number of holes,disappearing slit diaphragm in podocytes were observed in the model group.Compared to the model group,the pathological changes in podocytes were fewer in YYD-H group and the metformin.6.The protein expressions of nephrin and podocin: The protein expressions of nephrin and podocin in the control group were obviously highest among the all groups(P<0.05).The protein expressions of the model group were lower than the YYD-H group and the metformin group(P<0.05).Conclusion:1.The animal model of DN could be successfully established by SD male rats with high-fat diet accompanied by intraperitoneal injection of streptozotocin.2.Both YYD and metformin could improve the pathological changes in podocytes.The effect of YYD-H group was better than the YYD-L group and the YYD-M group.3.The mechanism of YYD's protective effect on podocytes in diabetic nephron-pathy rats may involves the upregulation of protein expression of nephrin and podocin.
