ObjectiveUsing metabolomics technologies to analysis the spectrum from chronic kidney disease stage 3-5 to patients with Qi deficiency of spleen and kidney or Yang deficiency of spleen and kidney, in order to explor potential Biomarkers of tranditional chinese medicine(TCM) syndromes and the inner material basis of TCM syndromes.MethodsThe study included 90 cases, of which 60 cases of non-dialysis patients with chronic kidney disease stage 3-5, of which 30 cases of i deficiency of spleen and kidney,30 cases of Yang deficiency of spleen and kidney; Moreover, 30 cases of healthy people were recrulited from Guangdong Provincial of TCM as control group.24-hour urine and blood were collected and tested. Serum was collected after high-Speed centrifugal blood, and saved them in-80? refrigerator. Using metabolomics technologies to analysis the spectrum in order to explor potential Biomarkers of tranditional chinese medicine(TCM) syndromes. Biomarkers'data were analysised by Markerview software and SIMCA-P 11.5 software. VIP(Variable Importance Projection) data were got after analysised by software. Top 100 compounds were choosed, and searched in the network database to determine the molecular formula.ResultsTraditional indicators, such as 24-hour proteinuria, renal function, liver function, blood lipid, fasting blood glucose, were no significant changes between Qi deficiency of spleen and kidney group and Yang deficiency of spleen and kidney group. Biomarkers of two tranditional chinese medicine (TCM) syndromes were Octadecanol?Capryloylcholine?9-Decenoylcholine? Phytosphingosine?Hexacosanoylcarnitine?12,13-DHOME?Tetradecanol??-glutamyl-L-putrescine?N-Decanoylglycine?Pimelylcarnitine 3,4-Dihydroxyphenylglycol?Ceramide (d18:1/20:0)?Terpenylisovalerate?Oleic acid?Petroselinic acid.Conelusion1. Traditional indicators were no significant changes between Qi deficiency of spleen and kidney group and Yang deficiency of spleen and kidney group, and new biomarkers were needed.2. Using metabolomics technology could indetify the differences between Qi deficiency of spleen syndrome and kidney or Yang deficiency of spleen and kidney syndrome clearly. Therefore metabolomics is a powerful tool for the researches of TCM syndrome differentiation.3. Metabolomics analysis suggests that metabolic differences between the two syndrome mainly exist in compounds such as amino acids, fatty acids, sphingolipids, and these compounds involved in fatty acid metabolism, lipid metabolism of amino acid metabolism, sheaths and linoleic acid metabolism. |
