ObjectiveThrough the establishment of negative emotional breast cancer-related fatigue animal model,exert the medium intensity vibration exercise intervention,through the tail suspension test and the immune indexes test,exploring the influence of vibration exercise on breast cancer-related fatigue in female rats with negative emotion as well as the change of the level of interleukin-1beta(IL-1 beta),tumor necrosis factor alpha(TNF-alpha)and interleukin 6(IL-6)before and after vibration exercise,in order to provide a scientific basis for the treatment of breast cancer-related fatigue of exercise and nursing plan,while exploring the IL-6 maybe plays an impact role in the processes.Methods1.Total of 70 healthy SD female rats were randomly divided into five groups: control group,CRF group,CRF+light group,CRF+noise group and CRF+light+noise group.Compared with the rats in control group of conventional breeding,the rats in other four groups were inoculated 0.2ml SHZ-88 breast cancer cells in their armpit,and then injected intraperitoneal epirubicin hydrochlotide of 2.5 mg/kg after successful inoculation,including two period of treatment.In the form of breast cancer cells inoculated and chemotherapy to finish breast cancer-related fatigue animal model.The rats in CRF+light group,CRF+noise group and CRF+light+noise group were given light stimuli,noise stimuli and light joint noise stimuli respectively to complete negative emotional breast cancer-related fatigue animal model.2.After modeling,the rats in CRF group,CRF+light group,CRF+noise group and CRF+light+noise were performed moderate intensity vibration exercise,the vibration frequency was 25 Hz,vibration time was 30 min,1time/day,six days/week for 8 weeks.3.Before modeling,after modeling and after vibration exercise,every groups rats were measured the weight and the rat tail suspension test(TST)to evaluate the fatigue status of the rats.Before and after negative emotion stimuli,every groups rats were measured open field test to evaluate the negative emotion status of the rats.After modeling and after the 8-week vibration exercise intervention immediately,taken blood from medial angle of eye and abdominal aorta by chloral hydrate solution anesthesia,then 3000r/min centrifuge for 10 minutes,take on the clear liquid,with double antibody enzyme-linked immunosorbent assay(ELISA)to detect the levels of serum IL-1?,TNF-? and IL-6.Results1.The general condition: Before the modeling,the difference of the weigh of the rats between different groups was not statistically significant(P>0.05).After modeling,the weigh of the rats in other four groups were lower than those in control group(P<0.01),the difference was statistically significant.The weigh of the rats in CRF+light group,CRF+noise group and CRF+light+noise group were lower than those in CRF group(P<0.01),the difference was statistically significant.The weigh of the rats in CRF+light+noise group were lower than those in CRF+light group and CRF+noise group(P<0.01),the difference was statistically significant.After vibration exercise,the weigh of the rats in CRF group,CRF+light group,CRF+noise group and CRF+light+noise group were higher as compared with themselves before vibration exercise(P<0.01),the difference was statistically significant.2.Rat tail suspension test(TST): Before modeling,the difference of the immobility time of TST of the rats between different groups was not statistically significant(P>0.05).After modeling,the immobility time of TST of the rats in other groups were longer than those in control group(P<0.01),the difference was statistically significant.The immobility time of TST of the rats in CRF+light group,CRF+noise and CRF+light+noise group were longer than those in CRF group(P<0.01),the difference was statistically significant.The immobility time of TST of the rats in CRF+light+noise group were longer than those in CRF+light and CRF+noise group(P<0.01),the difference was statistically significant.After vibration exercise,the immobility time of TST of the rats in CRF group,CRF+light group,CRF+noise group and CRF+light+noise group were shorter as compared with themselves before vibration exercise(P<0.01),the difference was statistically significant.3.Open field test: Before negative emotional stimuli,the difference of residence time,cross time and erecting time of the rats between different groups were not statistically significant(P>0.05).After negative emotional stimuli,compared with the control group,the difference of residence time,cross times and erecting time of the rats in CRF group were not statistically significant(P>0.05).Residence time of the rats in CRF+light group,CRF+noise group and CRF+light+noise group were higher and cross times and erecting time of the rats were lower than those in CRF group(P<0.01),the difference was statistically significant.Residence time of the rats in CRF+light+noise group were higher and cross times and erecting time of the rats were lower than those in CRF+light and CRF+noise group(P<0.01),the difference was statistically significant.4.The levels of serum IL-1?,TNF-? and IL-6: After modeling,the serum levels of IL-1? and TNF-? in CRF group,CRF+light group,CRF+noise and CRF+light+noise group were higher than those in control group(P<0.01),the difference was statistically significant.The serum levels of IL-1? and TNF-? in CRF+light group,CRF+noise and CRF+light+noise group were higher than those in CRF group(P<0.01),the difference was statistically significant.The serum levels of IL-1? and TNF-? in CRF+light+noise group were higher than those in CRF+light group and CRF+noise group(P<0.01),the difference was statistically significant.The difference of the serum levels of IL-6 between different groups was not statistically significant(P>0.05).After vibration exercise,the serum levels of IL-1? and TNF-? in CRF group,CRF+light group,CRF+noise and CRF+light+noise group were lower and the the serum levels of IL-6 were higher as compared with themselves before vibration exercise(P<0.01),the difference was statistically significant.Conclusions1.After inoculating SHZ-88 breast cancer cells and chemotherapy,the weigh of the rats were reduced and the immobility time of TST of the rats were longer,suggesting the degree of fatigue were aggravated.After the negative emotional stimuli,residence time of the rats were higher,cross times and erecting time of the rats were lower,suggesting negative emotion stimuli was succeed.In conclusion that negative emotional breast cancer-related fatigue animal model was established.2.After modeling,the fatigue degree of the rats accompanied by negative emotional stimuli were higher than pure fatigue rats,suggesting that negative emotion can increase breast cancer-related fatigue symptoms,and positively correlated with the severity of negative emotion.3.After vibration exercise,the levels of serum IL-1?,TNF-? were reduced and the levels of serum IL-6 were increased,the degree of cancer-related fatigue alleviated obviously,suggesting that vibration exercise can alleviate the CRF accompanied with negative emotion,and IL-6 maybe plays an impact role in the processes.
