The genus Ainsliaea,a member of the family Compositae,comprises approximately 70 species distributed mainly in south of Asia,44 of which are indigenous to China.The plants of the genus Ainsliaea,have antipyretic,haemostatic,anti-inflammatory and detoxifying properties,and have been long used as folk medicines in China,Korea and Japan.Phytochemical studies have indicated that this genus is a rich source of sesquiterpenes,and triterpenes.A.henryi,a folk medicinal herb growing mainly in Yunnan,Guizhou,Sichuan,Hunan,and Tibet provinces,and has been long used as a folk medicine for the treatment of traumatic injuries,edema,and fevers,colds,contusions.However,there is little phytochemical and biological studies of A.henryi have been reported to date.In this dissertation,we described the chemical constituents from A.henryi including their isolation,structural elucidation and bioactivities.Fifty five compounds were isolated from the whole plant of the A.henryi by various chromatography methods(silica gel,ODS,Sephadex LH-20,preparative-HPLC)and spectroscop analysis(~1H NMR,13 C NMR,~1H-~1H COSY,HSQC,HMBC,NOESY,HRESIMS,ESIMS,IR,UV),including twenty nine sesquiterpenoids,two diterpenoids,ten triterpenoids,two flavones,one steroids,and eleven others.Among these isolates,compounds AH-1,AH-12,AH-22,AH-32,AH-33,AH-34,AH-35,AH-46 were identified to be new compounds.Part of compounds were evaluated for cytotoxicity activity against four human tumor cell lines(A549,HCT116,MGC803,and CCRF-CEM)by MTT assay.Compounds AH-20,AH-34,AH-35,AH-38 and AH-55 showed strong cytotoxicity activities against four human tumor cell lines,while the others are weak or inactive.Besids,some sesquiterpenoids were tested for inhibitory activity agais LPS-induced nitic oxide(NO)production in RAW264.7 macrophages,and several compounds showed a moderate inhibition against NO release.
