Rehmannia henryi belongs to Rehmannia(scrophulariaceae),The chemical constituents of R.henryi whole plant water extracts were systematically studied by various chromatographic methods.The structure of the isolates were elucidated on the basis of spectral analysis and chemical evidence.Some of them were tested for their biological activity.88 compounds were obtained from aqueous extract of the whole plants of R.henryi.These compounds were identified as follows:jiohenrins A-P(1*,2*,4*,7-19*),henryioside A(3*),jiofuran carboxdehydes A-B(5*-6*),henryionosides A-B(20*-21*),4S-ethoxy rehmapicrogenin(22*),henryiamide A(23*),ajugol(24),dihydrocatalpol(25),catalpol(26),6-O-E-caffeoyl ajugol(27),rehmaglutoside F(28),aeginetoyl ajugol 5"-O-?-D-quinovoside(29),rehmaglutin B(30),rehmaglutin D(31),jiofuran(32),jiofuran aldehyde(33),jioglutolide(34),rhemaglutin C(35),rehmaglutin G(36),piscrosin E(37),dihydroxycatalpolgenin(38),rehmap icro genin(39),(4S)-rehmapicrogenin(40),rehmapicrogenin calcium salt(41),(4S)-rehmapicrogenin calcium salt(42),rehmapicrogenin sodium salt(43),frehmaglutoside A sodium salt(44),frehmaglutoside A calcium salt(45),(4S)-frehmaglutoside A calcium salt(46),rehmapicroside sodium salt(47),dihydroxy-?-ionone(48),aeginetic acid(49),aeginetic acid sodium salt(50),14-hydroxyaeginetic acid(51),rehmaionoside B(52),rehmaionoside A(53),(9R)-oxyrehmaio no side B(54),oxyrehmaionoside B(55),melasmoside(56),neo-rehmannioside(57),aeginetic acid 5-O-?-D-quinovoside(58),aeginetic acid 5-O-?-D-quinovoside sodium salt(59),aeginetic acid 5-O-?-D-quinovoside calcium salt(60),jiocarotenoside A1(61),jiocarotenoside A1 sodium salt(62),jiocarotenoside A1 calcium salt(63),4-hydroxyl-3-methoxybenzyl alcohol(64),3-hydroxyl-4-methoxybenzyl alcohol(65),3,4-dihydroxyphenylethanol(66),4-hydro xyphenylethanol(67),3,4-dihydroxyphenyl alcohol-?-D-glucopyranoside(68),2-(3,4-dihydroxyphenyl)-2(S)-hydro-xyethyl-3-O-?-L-rhamnopyranosyl-(1?3)-O-?-D-glucopyranoside(69),martynoside(70),isomartynoside(71),3,4-dihydroxy-?-phenethyl-O-?-L-rhamnopyranosyl-(1?3)-O-?-D-galacopyranosyl-(1?6)-4-O-caffeoyl1-?-D-glucopyranoside(72),jionoside A1(73),join-oside B1(74),p-coumalic acid(75),3,4-dihydroxyphenylpropanoic acid(76),ferulic acid(77),ethyl coumalate(78),syringic acid(79),3-hydroxy-4-methoxybenzoic acid(80),uracil(81),(+)-neo-olivil(82),stachyose(83),glucose(84),fructose(85),5-hydro-xypyrrolidin-2-one(86),ethyl-a-L-rhamnopyranoside(87),ethyl-a-D-galctopyranoside(88).Among the isolated eighty-eight compouds,there were thirty-four iridoids and iridoid glycosides(1*-19*,24-38),twenty-eight monoterpenes and sesquiterpenes(20*-22*,39-63),eleven phenethylalcohol glycosides(64-74),fifteen other components(23*,75-88).Twenty-three compounds(1*-23*)were new compounds.The cytotoxicity of 83 compounds obtained from the whole plant of R.henryi against 5 human tumor cell lines was tested by the MTT method.However,it was inactive for all cell lines used(IC50>10 ?M was defined as "inactive").Compounds 1*,2*,16*,19*,20*,22*,28-30,33,36-39,41-46,49,51,54-55,59,63,74 and 78-81 exhibited moderate hepatoprotective activities against APAP-induced HepG2 cell damage.The protective effect of the isolated compounds on the neurotoxicity induced by serum deprivation of PC 12 cells was investigated by MTT assay,and there was no obvious protective activities against neurotoxicity induced by serum deprivation in PC 12 cells.Compound 48 possesed obvious inhibition agaist the release of inflammatory factors of mice glia cell.For anti-diabetic biological activity,compound 74 has a significant inhibitory effect on aldose reductase with IC 50 values less than 7.5×10-6?M,and exhibited weak activities in inhibiting FBPasel,a-glucosidase,lipidase and DPP-IV enzymatic activity.
