Objective:The research was conducted to study the effects of 17?-E2 on gene n m23-H1?CD44 expression in endometrial stromal cells of patient with Endometriosis,and in an attempt to have a further study on the pathogenesis of endometriosis.Methods:The isolation and culture of endometrial stromal cells in vitro were coll ected from 9 patients with endometriosis having hysterectomy surgery.The cells were tr eated with 17?-E2 for different concentrations(0,10-8,10-6,10-4mmol/L),Trizol method was used to extract RNA from cells,and reverse transcription was used to produce cDN A,Real-time quantitative PCR was performed to determine the mRNA expression level of nm23-H1 and CD44 in the ESCs.Results:General qRT-PCR was adopted to have a test on the expression of nm23-H1 and CD44 in endometriosis matrix cells stimulated by 17?-E2 in different concentr ations.?1?The expression level of nm23-H1 in four concentrations(0,10-8,10-6,10-4mm ol/L)of stimulated stromal cells were 1.60±1.05,1.48±0.89,1.32±0.91,1.02±0.79 resp ectively,there was no statistical difference between the four groups(F=0.569,P>0.05.?2?The expression level of CD44 in four concentrations(0,10-8,10-6,10-4mmol/L)of sti mulated stromal cells were 2.18±2.14,3.39±2.93,2.79±2.61,4.51±3.92 respectively,th ere was no statistical difference between the four groups?F=1.004,P>0.05?.Conclusion:1.The expression of nm23-H1 decreased with the increasing of estrogen concentration in EM,indicating that estrogen may influence the existence and development of EM by acting on nm23-H1.The results were no statistical significance,which may due to the small number of samples in the experiment.The next step is to increase the sample number to make clear the relationship between nm23-H1 and EM.2.Estrogen has no obvious effect on the expression of CD44 in EM.
