A Research About Effects Of Bushen Huoxue Decoction On Lumbar Intervertebral Disc P38MAPKa Protein Level In Rat Without Ovary

Objectives: Observe the nucleus pulposus structure and pathological morphologic changes of degenerative lumbar intervertebral disc, and determine lumbar intervertebral disc p38 MAPK protein level in rat without ovary to investigate the effect of Bushen Huoxue decoction on the expression of lumbar intervertebral disc P38 MAPK and provide a former study for a further study on p38 MAPK signal path.What’s more,I will explain parts of mechanism of effect of Bushen Huoxue decoction on delay lumbar intervertabal disc’s degeneration based on my further study.Methods: The model was established by 60 healthy female SD adult mouse who is 12 month old and clean. Then they were divided into 5 groups randomly, each with 12 mouse, including false model group 、model group、 model 、model of TCM group and positive drug control group. Except for blank group and false group,all other groups mouse were used to make model of animal with osteoporosis after menopause. A week later,when all model were made successfully, they received different therapy with appropriate dose. The mouse of blank group,false model group and model group were feed on normal saline NS. Model of TCM group was feed with Bushen Huoxue decoction. except that,all groups were fed normally and weight them every 10 days, change the medicine dose according to their weight and lasted for 12 weeks. I picked 6 big mice randomly,killed them 4 hours after feed with medicine in the sixth weeks and eighth weeks, and then froze for biochemical tests.Result:Western Blotting test show that lumbar intervertebral disc p38 MAPK protein level of model group is higher than blank model and false model.And the differences has statistical significance(p<0.05).And it also proved the model of degenerative lumbar intervertebral disc and p38 MAPK protein level will increase as degeneration of lumbar intervertebral disc and the differences of p38 MAPK protein level in blank model and false model has no statistical significance(p>0.05).The p38 MAPK protein level in model of TCM group id lower than model group,thus its differences has statistical significance(p<0.05), so it indicate Bushen Huoxue decoction can reduce the expression of p38 MAPK protein and delay the degeneration of lumbar intervertebral disc.。the differences between Model of TCM group and positive drugs in p38 MAPK protein level has no statistical significance(p>0.05).Conclusion:1,the nucleus pulposus structure and pathological morphologic changes of degenerative lumbar intervertebral disc is the same as human.2,After I fed the adult mice after menopause with Bushen Huoxue decoction,lumbar intervertebral disc P38 MAPK protein level decreased obviously,which provides Bushen Huoxue decoction has delay the degeneration of lumbar intervertebral disc through delaying the degeneration of lumbar intervertebral disc’s structure and inhibiting the expression of P38 MAPK protein.
