Study On The Mechanism Of Yanggan Roujin Prescription Treats Achilles Tendon Enthesiopathy Based On COX-2/PGE-2 Pathway

Objective Through the method of "electric shock jump" to establish the model of rat achilles tendon enthesiopathy.Then treat the model rats by using "Yanggan Roujin" prescription.Observing the the behavior of rats,changes in cell morphology of terminal region,expression of COX-2/PGE-2 and the fiber morphological changes to explore the mechanisiam of Yanggan Roujin prescription on enthesiopathy.Methods 10 rats were randomly selected as the blank group,after established the model,the remaining 40 rats were randomly divided into 3 groups: "Yanggan Roujin" high dose group(high dose group), "Yanggan Roujin" low dose group(low dose group)and model group,the high dose group and low dose group use traditional chinese medicine to fill the stomach,simultaneously model group use physiological saline to fill the stomach.After the rats were anesthetized,cut the terminal region of the aehille tendon.After HE staining,observe the changes in cell morphology of terminal region.Detect the expression of COX-2 and PGE-2 by the method of ELISA,Observe the fiber morphological changes by transmission electron microscope.All datas were analyzed by SPSS 17.0.Results Pathological changes occurred after established the model,and the expression of COX-2 and PGE-2 was increased.After treatment, the high dose group and low dose group,compared with the model group,has largely improved in the pathology and the expression of COX-2 and PGE-2 has reduced,and the high dose group was more close to the blank group.Conclusion The model of rat achilles tendon enthesiopathy was successfully established in 8 weeks by the method of "electric shock jump". "Yanggan Roujin" prescription can promote the enthesiopathy recovery.The expression of COX-2 and PGE-2 in the terminal area may be one of the mechanisms of treatment.
