Objective:To establish the Enthesiopathy rat model through the "electric shock jump" training in 8 weeks. Observation the pathological changes and the expression changes of the IL-1β, IL-6, iNOS in terminal region of needle treated and control rats, To approach the mechanism of Enthesiopathy.Method:88 one-month-old male SD rats were randomly divided into normal group with 8, model group width 80. Model group give an "electric shock jump" training. After 8 weeks training,the model group were randomly divided into once treatment group with 64; four treatment group (1/3 days) with16.After 8 weeks, give a needle treatment to once treatment group, four treatment group at tendon pain points respectively. Then once treatment group were sacrificed after one,two,three,four days and the corresponding control group were sacrificed in the same day. After the fourth treatment of two days later the four treatment group put to death. HE staining to observe the terminal region morphological changes of cells and tissues. By immunohistochemistry to detect the expression changes of IL-1β, IL-6, iNOS in terminal region.Results:1. Model group have been considerable differences in morphology, phenomenon of disease symptoms, such as tendon fibers vague, disordered; "tide" apparent irregular contour of the "rising tide" phenomenon; calcified cartilage zone was significantly thicker than normal group.2. By HE staining microscopic observation:once needle treatment group had improved symptoms compared with the control group, four needle treatment group was more obvious than once needle treatment group.3. Between treatment of group 1,2 and control group the content of IL-1βexpression has no significant difference, the treatment group 3,4 both have significantly decreased IL-1β, while the treatment group 4 calcaneal articular surface expression of IL-1βwas significantly lower than the control group (P=0.03). The treatment group 4 and control group there was a marked decline in all regions, especially in the achilles tendon area (P= 0.036) there are significant differences.4.In treatment group 1 the expression of IL-6 levels lower than the control group in achilles tendon, fibrocartilage zone, but not statistically significant, treatment group 2, the IL-6 levels of fibrocartilage zone, calcaneal articular surface rise somewhat compared with the control treatment group, Treatment group 3 terminal region IL-6 levels decreased significantly, in the treatment group 4 fibrocartilage zone IL-6 decreased significantly, (P= 0.032). The treatment group and control group 5 there was a marked decline in all regions, especially in the calcaneal articular surface (P=0.045) there were significant differences in fibrocartilaginous zone (P=0.033) there are significant differences.5. treatment group 1 compared with the control group showed no significant change in iNOS expression,in the treatment group 2,3,the content of iNOS expression compared with the control group showed a downward trend, but without statistical significance, while in the treatment groups 4 the iNOS expression was significantly lower than the corresponding control group (P=0.024) in achilles tendon area. The treatment group 5 compared to control group various regions also have decreased significantly, especially in the achilles tendon (P=0.021), calcaneal articular surface (P=0.02) there are significant differences.Conclusions:1. "Electric shock jump" training can make Enthesiopathy model in 8 weeks.2. Once needle treatment can make Enthesiopathy improved, in particular, the tendon structure of peripheral tissue inflammation, from the view of the effect of four needle treatment, a reasonable increase in the number and the extension of needle treatment time there is a better effection.3. Once needle treatment can decrease inflammatory factors, a short time can delay the swelling. From the view of detection of inflammatory cytokines, four needle treatment group is more effective than one needle treatment group, but there is still the expression of various inflammatory cytokines, it is indicating that by increasing the number of needle treatment can ease the inflammation-terminal region,but enthesiopathy that requires a long recovery process.
