Abstract/Summary:
Objective: High-voltage burns cause not only the thermal damage to the body,but also the current through the body at the same time,the electric fields on the body’s blood vessels and nerves of the various groups have different degrees of damage.High-voltage after burn for circulatory system research has been carried out widely.Research shows that electrical burns after endothelial injury,such as local and systemic inflammatory reaction caused by hemodynamic changes,and thrombosis in microcirculation disorder.After electrical burns associated with systemic inflammatory response and hemodynamic change factors are many,including plasma adiponectin,fiber protease in regulating the inflammatory response and have great effect on the blood flow dynamics.Adiponectin(Adiponectin/ADP)is one of the types of fat cells endogenous bioactive peptides or proteins,which in endocrine circulating in the bloodstream,involved in regulating glucose and fatty acid metabolism and resisting inflammation and other life activities.Fiber protease(Brinase)is extracted from aspergillus oryzae proteinase,the fibrin can break down blood clots and reduce the plasma concentrations of fibrinolytic enzyme inhibitor.Electrical burns for fiber protease in the serum of rats after qualitative and quantitative change,there is no correlation research.This study by detecting high pressure point burn rats after the change of serum adiponectin,fiber protease,and through the Ulinastatin(Ulinastatin,UTI)intervention,to further explore serum adiponectin,fiber protease after high voltage change and its mediated inflammatory reaction and the effect of blood coagulation mechanism and intervention measures.Methods:1 Group: This experiment adopts the healthy adult SD rats as experimental study,the rats are provided by animal experiment center of hebei medical university(certificate no.1411023),a total of 180,according to randomized block design method which can be divided into three groups,each group had 60 rats respectively,such as false high voltage burn group(control group)(60),high voltage burns group electrical injury(group)(60),high voltage burns ulinastatin(UTI)in treatment group(treatment group)(60).Each group according to the length of the observation time again in six phases during different groups,namely the electrical injury before 15 min,5 min after electrical injury,electric injury after 1 h,2 h,4 h,8 h six groups,each group contains a phase 10 rats.2 Preparation:The rats were weighed and numbered.Disposition of experimental drugs to reach the required concentration.prepare blood collection needle,record the experimental data.3 Duplicate the high voltage electric burn model:The transformer and regulator wire connection,make it become a closed loop.After 10% chloral hydrate(0.3ml / 100g)intraperitoneal injection of anesthesia in experimental rats,the success of anesthesia,the rat limbs fixed,supine in the special shock experiments on the stage,depilating area of the(current entrance)of rat’s left arm and right lower extremity(current export)hair removal zone are respectively connected with the two 1 cm x 1 cm electrode.Ensure that all laboratory personnel from electric shock can be spread to the region,then connected to the power supply,a regulating knob,the output voltage of the voltage booster to 2 kV,turn on the power switch,high voltage current through the body of the rat,reach the shock effect,electric shock time is 3S,release of closed circuit within the residual current,rat cage and put it,to be blood.The blank control group(sham Electric Group)is not connected to the high voltage current,the remaining steps are the same as the electrical injury group and treatment group.After electrical injury group within 5 min in the abdominal cavity of rats injected with saline 5ml / kg,ulinastatin treatment group after 5min in rat peritoneal injection of 1% UTI(5ml / kg).Each sample is on time for blood,serum separation.4 Collection and detection: Operate the rat model which are replicated successful,open the chests,exposure heart,collect open-heart blood 6ml,at asepsis plastic tube,and let rest for 30 min,wait after serum precipitation,put the serums in centrifuge,to 3000r/min,centrifuge 10 min,take clear liquid in Eppendorf tube,preserve them in-70 conditions.℃5 Index test: By ELISA double sandwich antibody method,each group of rats were testing phase to six groupADP、Brinase serum content.6 Analysis of Statistics: Using SPSS 13.0 statistical software to finish variance analysis of two factors factorial design.It has significant statistically meaning if P<0.05.Results:1 Rats ADP serum content changesADP contents in Experiment groups are generally lower than those in control groups(ADP main effect F=17.336,P<0.05);ADP contents in experiment groups after injury are affected of time variable effect(the main effect F=20.094,P<0.05),after electric injury,from 5min to 4h,the level in each phase is lower than the level of pre-injury(P < 0.05),and is gradually heightening the trend,in the 4h phase to the lowest,and began to increase in the level of serum adiponectin in 4-8h,but the electrical injury group adiponectin level is still lower than the blank control group in corresponding time when adiponectin levels.ADP contents in UTI treatment groups are generally higher than those in the experiment groups(the main effect F=5.419,P<0.05);ADP contents in UTI treatment groups after injury are affected of time variable effect(the main effect F =20.708,P< 0.05),in the treatment groups after injury,from 5min to 4h,the level of ADP contents in each phase is lower than the value before the injury(P < 0.05).2 Rats Brinase serum content changesBrinase contents in electrical injury groups are generally higher than those in the control group(the main effect F =60.241,P < 0.05);Brinase contents in experiment groups are affected of tie variable effect(the main effect F =59.298,P < 0.05),after electric injury,from 1h to 8h,the level of brinase content in each phase is higher than the pre-injury level(P < 0.05),and is gradually heightening the trend.Brinase contents in UTI treatment groups is generally lower than those in experiment groups(main effect F =34.613,P < 0.05);Brinase contents in UTI treatment groups after injury are affected of time variable effect(the main effect F =81.736,P < 0.05),in the treatment groups after injury,from 1h to 8h,the level of content in each phase is higher than the value before the injury(P < 0.05).Conclusions:The expression of high-voltage electric burn can affect the body of adiponectin and the brinase induce excessive inflammatory reaction,and that is a hypercoagulable state.After high-voltage electric burn,early treatment of UTI can lead to lower the expression of plasminogen and increased expression of adiponectin,UTI can inhibit the excessive inflammatory reaction and regulating the balance of coagulation,improve microcirculation,promote wound repair.
