Objective: In the electrical burns,the dangerous rise along with the increase of duration of exposure,voltage and current intensity.Except the immediacy electric injury,there are also some unknown progressive damage.Several studies show that microcirculation plays an important role in the progress of the occurrence and development of the progressive damage after electric burns.Microvascular damage and microflow obstacles were the key problems.Now,in the field of the change of serum enzyme activity after high voltage electricity,researches have focused on serum creatine kinase(CK)and creatine kinase isoenzyme(CK-MB)and so on.The studies were less about the activity of lactate dehydrogenase(LDH)and myeloperoxidase(MPO).Through animal experimental model,our study tested the LDH and MPO of the high voltage burned rats and interferenced the experiment by Ulinastatin(UTI).The aims are to explore the characteristics of high voltage injury and to study the feasibility of UTI in the treatment of high voltage injury.Method:1 Experimental animal grouping: 180 healthy adult male SD rats(Hebei medical university animal experiment center,number of qualitative qualification: 1411032)were divided into three groups according to random number table method.They were false high voltage burn group(control group),high voltage burn group(electrical injury group)and Ulinastatin group(UTI group).The number for each group is 60.Each group was divided into six subgroup according to the observation time which were 15 min before injured,5 min after injured and 1h,2h,4h,8h after injured.The rats in each subgroup were 10.2 The preparation before experiment: number and weigh the rats,denude the hair in left upper limb,right upper limb and chest.Make experimental drugs according to the required concentration.3 High voltage burn model: Intraperitoneal injection of 1% sodium pentobarbital was done for anesthesia(40mg/kg).After the success of the anesthetic,we put the rats on test bench.Two electrodes(1cm×1cm)were respectively fixed in the left upper limb(current entrance),the right lower limb(current export).The high voltage sustained for 3 seconds and voltage was 2kv.The control group only got the wire without electricity.Within 5 min after injury,intraperitoneal injection of 1% UTI was given in the UTI group(2mL/kg).The control group and electrical injury group were given the NS(2mL/kg).4 Specimen collection and preservation: Operate the rat model which are replicated successful,We draw blood for 4ml with an opened chest.And then,the blood are put in plastic tube and stewing for 30 min.Then,centrifuging the blood for 10 min with 3000r/min.The supernate was placed in Eppendorf tube and saved in 70 conditi? onsIndex exam: Through the ELISA ouble sandwich antibody method,each group of rats were testing phase to six group LDH and MPO serum content.5 Statistics: The experimental data obtained is measurement data which is described by mean and standard deviation.The SPSS 13.0 was used for processing data.variance analysis of two factors factorial design and LSD method t test method were applied.The test of significance level is p<0.01.Result:1 Changes of LDH in ratsCompare with control groups,LDH in experiment groups are generally higher(main effect F=304.321,P<0.01).They are affected by time variable effect(the main effect F=71.338,P <0.01).From 5min to 8h after electric injury,they present the trend of increasing(P < 0.01)LDH contents in UTI treatment groups are generally lower than those in the experiment groups(the main effect F=199.753,P <0.01);in UTI treatment groups,they are affected of time variable effect(the main effect F = 88.457,P < 0.01),,From 5min to 8h after injury in the treatment groups,the levesl of LDH are higher than the value before the injury(P < 0.01).2 Changes of MPO in ratsMPO contents in electrical injury groups are generally higher than those in the control group(the main effect F = 362.545,P < 0.01);MPO in experiment groups are affected of time variable effect(the main effect F = 112.350,P < 0.01).From 5min to 8h after electric injury,the levels of MPO in each phase are higher than before(P < 0.01).The trend is rised constantly.MPO contents in UTI treatment groups is generally lower than those in experiment groups(main effect F = 289.452,P < 0.01);MPO in UTI treatment groups after affected of time variable effect(the main effect F = 85.342,P < 0.01).From 5min to 8h after injury in the treatment groups,the levels of MPO in each phase are higher than the value before the injury(P < 0.01).Conclusions:1 LDH is increased in High-voltage electric burned rats,and it is gradually increasing2 MPO is increased in High-voltage electric burned rats,and it is gradually increasing3 High-voltage electricity promoted the inflammation by stimulating the production of LDH and MPO.They are also influencing the microcirculation.4 UTI could inhibit the production of LDH?MPO.So the inflammation and microcirculation disorder are alleviated...
