Objective: to SD rat traumatic femoral head necrosis model as the research object, detection of Taohong Siwu Decoction on effect of GPR48 expression in SD rat traumatic femoral head necrosis process, evaluation of Taohong Siwu Decoction to improve the avascular necrosis of the femoral bone blood supply and osteonecrosis of the new effect.Methods:Methods: by Hunan University of traditional Chinese medicine animal laboratory unified purchase of 29 SD rats, adaptive feeding 1 week after weighing, then according to the weight sequence number. Finally, according to the random number table method were divided into 3 groups, namely normal control group, sham operation group and model group, each with 8, 8, 13. According to the experimental method of Norman after modeling 4 weeks, from 3 SD rats were randomly selected from each group to begin testing the modeling success, after making a successful model in accordance with the principle of random will build the module were randomly assigned to two groups treatment group, model group, each group 5, the treatment group use Taohong Siwu Decoction by intragastric administration, other groups using equal volume of physiological saline was infused into the stomach, intragastric 8 weeks after, each group SD rat femoral head sampling and shall be put to death, sample submission, content detection GPR48 m RNA.Results:(1) gross specimen observation: the normal control group after modeling 4W, after intragastric administration of 8W rat articular cartilage of the femoral head surface is smooth, shiny, milky white. The sham operation group after the model 4W, after intragastric administration of 8W rat articular cartilage of the femoral head surface smooth, luster dim, is milky white. The model group 4W rat femoral head cartilage surface is yellowish white, slightly rough, eroded appear different degree of damage, after intragastric administration of 8W rat femoral head was dark red, rough surface of articular cartilage, articular cartilage surface layer have different degrees of collapse. The treatment group 4W rat femoral head cartilage roughly consistent with the model group, after intragastric administration of 8W rat femoral head cartilage surface is dark red, rough surface, cartilage collapse degree is lighter than the model group.(2) the X-ray results showed: normal control group, sham operation group after intragastric administration of 8W rat femoral head change is slight, the model group and treatment group irrigation stomach after 8W when density to reduce the area and bone trabecular derangement, different layer of the collapse of the articular cartilage surface.(3) by HE staining and Masson results show after modeling 4W, filling the stomach after 8W blank control group, sham operation group, the femoral head bone trabeculae arranged in neat rows, dense, medullary cavity empty, hematopoietic tissue normal, there are a small number of empty bone lacuna, model group, treatment 4W weeks necrosis focus in the subchondral bone and epiphysis cartilage zone, but also around the necrotic trabecula local active signs of bone cell, and a small amount of new bone formation, visible form a large number of empty bone lacuna, bone trabecula was sparse, disorder, fracture, collagen content significantly reduced. Irrigation stomach after 8W necrosis region to further expand and medullary cavity empty and hematopoietic tissue decreased significantly, a substantial reduction in trabecular bone cells, a large number of empty lacuna formed, the contour of the femoral head have different degrees of collapse, from each group can be found in model group, treatment group of trabecular bone in bone lacuna rate were significantly higher than those in the sham operation group and blank control group. Statistical significance(P < 0.001).(4) RT-PCR analysis results showed: blank control group, sham operation group, model group, GPR48m RNA expression gradually decreased, but the treatment group GPR48 m RNA expression compared with model group, the amount was slightly higher, and lower than that of the normal control group, sham operation group, the differences of statistical significance(P < 0.001)Conclusion: GPR48 rat traumatic femoral head necrosisdevelopment in the process of expression gradually decreased, but the Taohong Siwu Decoction stem prognostic GPR48 m RNA was slightly higher, but lower than the normal value, which may be related to the local repair of necrotic bone.
