objective: Through the observation of Chaishao gastritis granule(the English referred to as CSGG) on gastric ulcer in rat mucosa Bcl-2 and NF ?B protein positive expression, in order to further explore the possible principle and methods of the CSGG in the treatment of gastric ulcer, for clinical treatment of the disease by clinical and experimental study.Methods :Forty healthy Wistar rats, that 20 male and 20 female, body weight(190±40)g, were randomly divided into four groups according to, respectively, the normal control group, pathological model group, CSGG group and omeprazole group, 10 rats in esch group. Adaptive feeding for 7 days, then the pathological model group, CSGG group and omeprazole group were according to Takagi method of making acetic acid rat model of gastric ulcer. Modeling for 3 days, were treated with distilled water, omeprazole, Chaishao gastritis granule by gavage for 13 days to drug withdrawal. Then the rats were fasted 24 h and killed by cervical dislocation, except the control group, the other 3 groups measured ulcer index. The samples were fixed in 4% formalin for routine paraffin-embedded sections, and using immunohistochemical staining gastric mucosal cells Bcl-2, NF-?B protein positive density tests.Results:(1)The CSGG group and omeprazole group can significantly reduce the ulcer index of rats(P<0.01). Compare the CSGG group with the omeprazole group, the difference was statistically significant(P<0.01).(2)The positive expression of Bcl-2 by the CSGG group and omeprazole group was significantly higher than pathological model group(P<0.01),and the particle in CSGG group expressed stronger than that in omeprazole group(P<0.01).(3) The expression of NF-?B in CSGG group and omeprazole group was lower than the model group, the difference was obvious statistical significance(P<0.01). And the difference between CSGG group and omeprazole group was no significant difference(P>0.05).Conclusion: Chaishao gastritis granule can effectively improve the clinical symptoms of rats and reduce ulcer index, promote the healing of ulcer. Chaishao gastritis granule can enhance rat gastric mucosa Bcl-2 positive expression and plays a role in inhibiting apoptosis and further repair the damaged mucosa and to promote ulcer healing. The positive expression of NF-?B was positively correlated with the rat ulcer index. Chaishao gastritis granule can reduce the excessive inflammatory response and promote the healing of ulcer by inhibiting the activation and expression of NF-?B. NF-?B may be one of the factors that affect the healing of ulcers.
