Objective: to use with sun Jiao, treatment of alcoholic liver disease(alcoholic liver disease, ALD) experience prescription "gehuang Granule" on rat model of alcoholic fatty liver intervention, by observing the rat serum index changes and pathological changes, a preliminary study of Ge Huang granule in preventing and treating alcoholic fatty liver may be involved in the mechanism.Methods: selected 108 male and female SD rats, divided them for six groups: gehuang granule low dose group(low dose group), Ge Huang particles in the dosage group(middle dose group), particle Ge Huang high dose group(high dose group), the United States metadoxine in the positive control group(positive control group), model group and normal group(n = 18). The rat model of alcoholic fatty liver was prepared by the method of gradient liquor filling and high fat diet. The anhydrous ethanol disposition into 30%, 35% and 40% concentration, high, of medium, low, model group and positive control group in accordance with the 1.0ml.100g-1.d-1 dose respectively irrigation stomach 1, 1, 10 weeks. At the same time from the first weeks of the start to the end given every other day high fat diet and normal group were given the same volume of distilled water irrigation stomach and ate a normal diet. After 4 weeks of high, medium and low dose group were treated with high, middle and low dose of gehuang granule, while the control group was given the beauty of metadoxine in gavage. Lasts 8 weeks after sacrificed all the rats. HE staining was used to observe the pathological change in the liver tissue of rats; by using ELISA enzyme linked immunosorbent assay for the detection of IL-1 P, IL-6 and tumor necrosis factor(TNF alpha), alcohol metabolizing enzymes(ADH), to metabolize acetaldehyde dehydrogenase(ALDH), leptin(leptin(LP), adiponectin()); using tachypleus amebocyte lysate chromogenic matrix method for the detection of endotoxin lipopolysaccharide(LPS).Results: 1. rat liver tissue HE staining results: color uniform normal rat liver tissue and normal structure of hepatic lobules, hepatic cord along the central vein as the center of radial arrangement and clear outline of liver sinus; after 4W model group liver tissue structure remained intact, partial liver cell is partially visible small vacuoles, for mild fatty liver pathological features; 12 weeks of modeling in model group, hepatic tissue visible large fatty degeneration, liver tissue structure disarranged, fuzzy boundary of liver cells, liver cells visible a lot of fat vacuolization and liver nuclei is fat droplets extruded, tend to cell membrane. High dose group, positive control group, the basic recovery of the liver cell structure, the middle dose group compared with the model group has improved, the low dose group and the model group is basically similar to the pathological changes.2. The changes of serum ADH and ALDH indexes in rats: the expression of ADH was significantly increased in model group(P<0.05), and the serum ALDH content was significantly lower(P<0.05). After drug intervention ADH content in high dose group and positive control group decreased significantly(P < 0.05), medium and low dose group than those in the model group decreased obviously(P > 0.05), high dose group and the control group no significant difference(P > 0.05). ALDH in high dose group and positive control group were significantly increased(P<0.05), the middle and low dose group compared with model group was not significantly increased(P>0.05), high dose group and control group was no significant difference(P>0.05).3.rat serum IL-1 P, IL-6, leptin, adiponectin, and plasma endotoxin, TNF, index changes: in the model group rats serum IL-1 beta and IL-6, leptin and adiponectin levels in increased significantly(P < 0.05), after drug intervention, the index content in high dose group and positive control group decreased significantly(P < 0.05), medium and low dose group compared with the model group, the decline is not obvious(P > 0.05), between the high dose group and the control group no significant difference(P > 0.05). In the model group rats plasma endotoxin and TNF alpha levels significantly increased after drug intervention, endotoxin, TNF content content in high dose group and positive control group decreased significantly(P < 0.05), medium and low dose group than those in the model group decreased obviously(P > 0.05), high dose group and the control group no significant difference(P > 0.05).Conclusion: 1. The high fat diet and high fat diet were successfully established in the rat model of alcoholic liver disease.2.particles of Ge Huang can reduce the content of alcoholic liver disease in rats serum ADH and ALDH content increased, through the promotion of ethanol metabolism to prevent liver damage, thereby reducing the plasma concentration of ethanol, to effectively reduce the alcohol hangover effect, reduce the damage of alcohol on the body and liver in rats.3.particles of Ge Huang can decreased significantly the content of bacterial endotoxin in the rat model of alcoholic liver disease, and regulation of cytokines IL-1, IL-6, TNF alpha, leptin, adiponectin, inhibit the inflammation of the liver, reduce the pro-inflammatory cytokines, extracellular matrix etc. and deposition, blocking the development of pathological process of alcoholic liver disease.
