objective: In this paper,To study the effect and mechanism of Ge Huang Granule on animal model of alcoholic liver disease,to provide theoretical basis for clinical application and further development of the drug.Methods:110 male rats were selected,then randomly divided into 6groups,namely the normal control group(normal group),model group,metadoxine control group(control group),the high,middle and low dose groups of Ge Huang granule(referred to as high dose group,middle dose group and low dose group),The normal group was 10,and the other group was20.Model group,High,medium and low dose groups of Ge Huang granules and control group were given the The Red Star Erguotou gavage(The alcohol concentration was 30%-53%,and the dosage was 2.0ml.100g-1.d),the control group was given the same dose of distilled water,All animals were fed with ordinary feed.After 6 weeks of intragastric administration,drug intervention was initiated,gehuang granule groups were given different concentrations of gehuang granule solution by intragastric gavage,the control group of metadoxine solution.After 12 weeks of intragastric administration,all rats were killed,Pathological changes of liver tissue were observed by oil red O and HE staining;Detection of serum AST,ALT,TC,TG,Liver tissue TC,TG;Western Blot was used to detect the expression level of cytochrome P4502E1 in livercells;The indexes of alcohol metabolizing enzymes(ADH)and acetaldehyde metabolizing enzymes(ALDH)were determined.Results: 1.The results of O staining and HE staining of liver oil Oil red O staining:n normal group,the morphology of hepatocytes was normal,the structure was clear,and the nuclei were blue,and there were no obvious red lipid droplets in hepatocytes.In the model group,the liver cells of rats were significantly enlarged,and the diffuse red lipid droplets were formed in the visual field,some confluent into pieces,nuclear is lipid drops to one side.In the high,middle and low dose group,the red lipid droplets in liver cells were significantly lower than those in the model group,and with the increase of the dose,the red lipid droplets decreased gradually,then recovered to normal.Compared with the normal group,there was no significant change in the morphology of hepatocytes in the control group.HE staining:Normal group of rat liver cell structure is clear,border and clear,neat.the model group,the morphology of hepatocytes was significantly enlarged,the structure was not clear,the boundary was fuzzy,the arrangement of cells was disordered,and the fat vacuoles of different sizes were seen in liver cells,a few nucleus were pushed aside.The structure and morphology of the liver cells of the rats in the high dose group were significantly improved,and the liver cells were arranged in order,only a few cells lack of uniform size,mild swelling.Compared with the model group,the structure and morphology of the liver cells in the middle dose group were improved,but there were still some swelling,the low dose group showed no significant changes in pathology,the liver cells were still swollen,the arrangement of cells was disordered,and the fat vacuoles were found in liver cells.The control group structure of liver cells were arranged in order,only a handful of fat vacuole formation in liver cells.2、liver index、serum AST、ALT、TG、TC,liver tissue TG、TC,P4502E1 protein expression:The above indexes in model group were significantly higher(P<0.001);After the gehuang granule and metadoxine drug intervention,these indicators are high,medium and low dose group and decreased in varying degrees,the control group and the high dose group was significantly lower(P<0.001),the middle and low dose group was not significantly decreased,and there was no statistical significance(P>0.05);there was no significant difference in liver index,serum AST,ALT,TG,liver tissue TG,P4502E1 protein expression between the high dose group and the control group(P>0.05),but there was statistical significance in TC(P<0.05).3、serum ADH and ALDH content:Compared with the normal group,the content of ALDH in serum was significantly lower in the model group(P<0.001),the expression of ADH was significantly increased(P<0.001),after the gehuang granule and metadoxine drug intervention,ALDH in high dose group and control group were significantly increased(P<0.05),ADH in high dose group and the control group was significantly decreased(P<0.05),In the middle and low dose group,the increase of ALDH and the decrease of ADH were not significant,and there was no significant difference(P > 0.05);There was no significant difference between the high dose group and the control group(P >0.05).Conclusion:1.The simple gradient liquor can successfully establish the rat model of alcoholic liver disease.2.Ge Huang granule can improve the alcoholic liver disease rat liver pathology damage degree,reduce the accumulation of fat and blood lipid,improve liver function,and has good effect on ALD.3.Ge Huang granule can make the the alcoholic liver disease in rats serum ADH content decreased significantly,make ALDH content increases,It can make the metabolism of ethanol in the body to get fast degradation,reduce the retention time and the concentration of ethanol in blood plasma,thus play a role,protect liver therapy.4.Ge Huang granule can reduce the expression of P4502E1 protein,which indicates that it may be related to oxidative stress in the prevention and treatment of ALD.
