Fatty Liver (FLD) refers that the pathological changes caused by various reasonswhich excessive fat accumulate in the liver cells. There are many reasons can causeFLD, such as obesity fatty liver, alcoholic fatty liver, fast weight loss fatty liverdisease, diabetes fatty liver, drug-induced fatty liver, fatty liver of pregnancy and soon. FLD threat to people’s health seriously. Shenfei granule is a traditional Chinesemedicine which provided by Bethune Medical University Pharmaceutical, consistingof ginseng, radix codonopsis, hawthorn, milk thistle bing. And it is intend to developtraditional Chinese medicine six class of new drugs treatment of fatty liver. Thisarticle choice alcoholic and non-alcoholic fatty liver model in rats, and observe theprotective effect of Shenfei granule on alcoholic and non-alcoholic fatty liver disease,and provide pharmacological basis for the development of a new Chinese medicine.1The Protective Effect to AFLD150male Wistar rats, suitable after a week,12rats were normal group, the otherrats were made alcoholic amimal model:1-4weeks the rats were administered orallywith40%alcohol8g/kg,5-8weeks the rats were administered orally with50%ofwine9g/kg,9-16weeks the rats were administered orally with50%of wine9g/kg.From the ninth week the model rats were randomLy divided into five groupsaccording to the weight: model group, and Shenfei granule (1,2,4g/kg), SuiFeiJiBin(80mg/kg). From the ninth week the rats were administered orally correspondingdrugs, and normal group and model group were given to equal volume of0.5%sodium carboxymethyl cellulose. Rats were anesthetized by intraperitoneal injectionof chloral hydrate, then abdominal aortic and separated blood serum, Test serum TC,TG, VLDL, FFA, MDA levels and SOD, AST, ALT activitives; take some liver asHE; some were made homogenates to test TC, TG, MDA levels and SOD activity.Results: Compared with normal group, in model group the content of TC, TG,VLDL, FFA and MDA in serum increased significantly, and the activity of SOD decreased, and the activity of AST, ALT increased. MDA levels were significantlyhigher in liver homogenates, and the activity of SOD decreased, and liver indexincreased. Compared with the model group, Shenfei granule2,4g/kg dose group,could significantly reduce serum TC, TG, VLDL, MDA, FFA levels and ALT activity,and increased SOD activity. Shenfei granule2,4g/kg dose significantly reduced liverTC and TG levels, and the content of MDA in liver tissue was significantly lower inShenfei granule4g/kg dose group. Pathomorphological, Shenfei granule couldsignificantly improve the liver tissue lesions.2The Protective Effect to NAFLD100male Wistar rats, suitable after a week,12rats were normal group, the otherrats were gave a high-fat feed. From the fifth week, the model rats were randomLydivided into five groups according to the weight: normal model group, and Shenfeigranule (1,2,4g/kg), SuiFeiJiBin (80mg/kg). And the rats were administered orallycorresponding drugs, and normal group and model group were given to equal volumeof0.5%sodium carboxymethyl cellulose for continuous4weeks. After4weeks allthe rats were anesthetized by intraperitoneal injection of chloral hydrate, thenabdominal aortic and separated blood serum, Test serum TC, TG, LDL-c, HDL-c,VLDL, FFA, MDA, NF-κB, TNF-α, IL-6levels and SOD, AST, ALT, CHE, LDHactivitives; take some liver as HE; some were made homogenates to test TC, TG,MDA levels and SOD activity.Results: Compared with normal group, in model group the content of TC, TG,MDA, NF-κB, TNF-α and IL-6in serum were significantly elevated. HDL-c levelsdecreased significantly and ALT activity elevated. MDA content in liver homogenatessignificantly increased, and SOD activity decreased, and liver index increasedsignificantly, and VLDL, CHE, LDH and AST did not change significantly.Compared with model group, Shenfei granule4g/kg dose could significantly reduceserum TC, IL-6levels and ALT activity; Shenfei granule2,4g/kg dose couldsignificantly reduce serum TNF-α, NF-κB, TG levels, and increased HDL-c levels andSOD activity, and decreased ALT activity; Shenfei granule1,2,4g/kg dose significantly reduced MDA levels in serum; Shenfei granule4g/kg dose groupsignificantly reduced liver TC levels, and increased SOD activity; Liver tissue MDAand TG content were significantly lower in Shenfei granule2,4g/kg dose group.Pathomorphological, Shenfei granule2,4g/kg dose could significantly improve theliver tissue lesions.
