Actinomycetes are well-known for their extraordinary ability to encode secondary metabolites,and are a type of microbial resource with enormous application value.With the increasing difficulty of excavating new actinomycete resources from the general environment,many scientists have focused their attention on primitive habitats that have not been disturbed by human activities.The Qinling Mountains are the dividing line between our country's northern and southern geography and climate.They have large areas of virgin forests,and huge mountain gaps form their unique vertical climatic zones.The vegetation belts and soil types also change accordingly and contain abundant actinomycetes resources.In this study,the forest soil samples were collected from different altitudes and different vegetation zones on the northern and southern slopes of the main peak of the Qinling Mountains.The physicochemical properties of soil samples with different slope direction,elevation and vegetation types were studied systematically.The actinomycetes were separated by designing different media.The number and types of actinomycetes were counted,the ecological distribution of actinomycetes was analyzed,and the influence of soil physical and chemical properties,elevation,slope direction and forest land types on the number and types of actinomycetes were analyzed;A new specie of actinomycete were identified by multiphase classification.The main findings are as follows:1.The pH change range was 5.60-6.41 on the southern slope and 5.07-6.58 on the northern slope.The change range on the northern slope was greater than that on the south slope;the change range of the organic matter content on the northern slope was greater than that on the southern slope,and the differences in organic matter content between the various vegetation belts on the southern and northern slopes.The content of organic matter of Abies fargesii is relatively high,and the content of soil organic matter is the lowest in theQuercus aliena;the change ranges of ammonium nitrogen content on the north and south slopes are similar,the content of ammonium nitrogen in the soil samples of the sharp-toothed alfalfa is the lowest,and the content of nitrate nitrogen in the north and south slopes is the lowest.The trend of elevation with altitude increased first and then decreased,but there was no correspondence between the north and south slopes.The highest nitrate content in the northern slope was Rhododendron purdomii,while in the southern slope wasAbies fargesii.he content of available phosphorus in the northern and southern slopes is relatively high in the soil samples of Larix chinensis,while the content of available potassium in the mixed forests of Populus alba and Pinus sylvestris is the lowest.It is 2.20 mg/kg.There is no obvious trend of available potassium content with altitude,but the lowest available potassium content is in the Rhododendron capitatum Maxim with the highest altitude on the north and south slopes.2.The number of actinomycetes changes in the direction of the middle high and low,that is a large number of low-altitude,a small number of high altitude.Low-altitude Sharp Tinea Philoxeroides have obvious advantages in the number of actinomycetes,and the number of actinomycetes in the high altitude Alpine Shrubs and Alpine Grass was the lowest;38 genus of actinomycetes were isolated from 41 samples,a total of 164 species,of which the majority of actinomycetes belong to the genus Streptomyces,with a total of 92 species,while Nocardia,Amycolatopsis,and Kitasatospora are predominant in the other actinomycetes,while the other 28 Actinomyces genus isolates only one strainactinomycete,such as Moraxella,Micromonospora,and Nocardiopsis;the abundance of actinomycetes and genera in soil samples from the northern slope is higher than that in the southern slope.In the comparison of vegetation types,the abundance of actinomycetes and genus isolated from pure forests of Quercus acutidentata was the highest,and the endemic genus was the most,but the number of strains of high altitude Alpine Meadows and Alpine Shrubs,and the number of actinomycete and endemic genus are the least.3.The quantity of actinomycete was significantly negatively correlated with organic matter and available potassium,and it was also negatively correlated with other physical and chemical properties.There was no corresponding relationship between the differences in the quantity distribution of actinomycetes and the differences in physical and chemical properties between the samples.The endemic genus of the low altitude region belongs to the Kribbella and Leifsonia.There are 15 and 30 endemic strains to the northern and southern slope of the low-altitude area,accounting for 37.50% and 54.55% of the total strains on the northern and southern slopes,respectively.The endemic strains of Quercus acutidentata and mixed forest of Pinus armandii Franch and Populus purdomii Rehd were 19 strains and 26 strains respectively,accounting for 44.18% and 52.00% of the total strains respectively;the endemic genus in the middle altitude area was Nocardia and Micromonospora and Paenarthrobacter,and the endemic strains of the northern and southern slopes were 12 and 13 strains respectively,which account for 40.00% and 41.94% of the total strains on the northern and southern slopes.13 strains and 21 strains The endemic strains of betula utilis and abies fargesii are 13 and 21 strains respectively.And the endemic genus in the high altitude region belonged to Curtobacterium and Mycobacterium;9 strains were endemic to both the north and the south,which accounted for 33.33% of the total strains on the south and north slopes,respectively.There were 11 and 17 strains of endemic strains in the Larix chinensis Beissn and Rhododendron capitatum,accounting for 45.83% and 56.67% of the total strains,respectively.In the three altitude,elevation,slope direction and vegetation type all have an impact on the ecological distribution of actinomycetes in soil.4 A new species of potential novel actinomycete obtained through 16 S rRNA cloning and sequencing analysis,and it was found that its similarity with Amycolatopsis xylanica CPCC 202699 was the highest,and that with other Amycolatopsis strains was less than 97%.Through the morphological characteristics,physiological and biochemical indicators,chemical indicators and DNA-DNA hybridization assays,it was confirmed that AFM 30020 is a novel species of Amycolatopsis.
