Phytolaccae The Extraction Of Active Ingredients And Its Effect On The Control Of Chicken Kidney Disease Of Research

Objective:Phytolaccae belongs to completed by water medicine,which has been widely pharmacological effects,this study aims to further explore its resisting nephritis,screening of swollen chicken kidney disease treatment effect is the best extraction method,for the guidance of clinical use and research and development of new drugs provide a scientific basis.Method:(1)Decoction method of water extract-alcohol precipitation and the method(final concentration of alcohol precipitation were 60%,70%,80%,90%),decompression drying,diacolation method(ethanol concentration were 50%,70%,90%)4 kinds of methods for extracting the effective component in the Phytolaccae,mainly compare the content of the extraction method of EsA.(2)Chicken kidney swelling model,then the treatment with the extracts of Phytolaccae,and setting up of veterinary drug control group and positive control group,were observed before and after treatment between groups of chicken clinical symptoms,body weight changes,cured,viscera index,serum uric acid,creatinine and urea,kidney tissues NO content,interleukin 2 receptor level and Caspase 3 activity.(3)The Phytolaccae diacolation extracts the curative effect of treating swollen chicken kidney disease to carry on the safety evaluation,experimental animals 5 groups,respectively,the normal dose group,normal dose three times,five times,ten times group and the blank control group,observed experimental animals during the medication,the serum biochemical indicators and clinical symptoms,hematology indexes have tissue biopsies compared with blank control group without significant difference.(4)The EsA content determination and thin layer identification about the methodology of specificity,repeatability test were studied.Results:(1)Experimental results show that with 10 times the amount of 90% ethanol extraction of 12 h,the rate of 1 mL/min diacolation and get the highest content of EsA and the highest withdrawal rate,respectively1.72 mg/mL and 49.14%.(2)Clinical pharmacodynamics research results showed that different methods to extract the Phytolaccae extracts of swollen chicken kidney disease have significant improvement and treatment effect,of which the Phytolaccae diacolation extract the best treatment effect,can significantly reduce the model set of chicken serum UA,Cr,Urea,kidney tissues NO,interleukin 2 receptor level,there was no significant difference between the control group and the control group(P>0.05),There was no significant difference in Caspase-3 activity between the model group and the control group(P>0.05).(3)Chicken kidney swollen Phytolaccae diacolation extract treated safety evaluation experiment results show that: during the clinical symptoms of treatment group has no obvious change,Phytolaccae diacolation extract the highest dose group weight at the end of the medication period is lower than the blank control group,but no significant difference(P > 0.05),the treatment group hematology index and serum biochemical index there was no significant difference compared with the blank control group,not seen obvious abnormity autopsy and histological observation.(4)Quality research results show that,the establishment of EsA detection method of strong specificity,high repeatability.Conclusion:Study with 10 times the amount of 90% ethanol extraction of 12 h,the rate of 1 mL/min diacolation and get the highest content of EsA and the highest withdrawal rate,respectively1.72 mg/mL and 49.14%,and in the stu dy of pharmacodynamics,improve and treatment effect of kidney enlargement is most prominent,Es A is likely to be the treatment of kidney disease one of th e effective ingredients of swollen;In the safety evaluation of each treatment gr oup hematology indexes and serum biochemical indexes compared with blank c ontrol group no significant difference,autopsy and histology observation not see n obvious abnormity,illustrate the Phytolaccae extract for the treatment of kidn ey disease of chicken is safe and effective;Finally established the Es A content determination and thin layer identification method,the method is simple and eas y to operate,stable and reliable,provide the basis for better control quality.
