Bovine mastitis is a pathological process of inflammation in mammary tissue,and a common disease,which represents complex causes.It not only leads to decrease milk production and milk quality but also increases the cost of treatment.In serious cases,it will cause the culls and death of dairy cows.Therefore,bovine mastitis caused huge economic losses to the dairy industry.Difutongru oral solution,composed of Dandelion and Polygalae radix,et.al,has been proved that it has a good clinical effect to treat bovine mastitis.The following experiments were carried out in this study:production process optimization,anti-inflammatory test,bacteriostasis experiment in vitro,and clinical efficacy verification et.al.1.L16(44)orthogonal test was designed to optimize the liquid-material ratio,boiling time,and extraction times of Difutongru oral solution.The optimal extraction process A1B2C3 was determined through the yield of dry extract and the content of total phenolic acid,in which the weights were set as0.6 and 0.4,respectively.A1B2C3 was 8 times of liquid-material ratio,1 h of boiling time,and 3 times of extraction times.2.Xylene auricle inflammation test and acetic acid capillary permeability test in mice were used to investigate the inhibitory effect of Difutongru on acute inflammation.The results showed that auricle swelling degree in the high-dose group was significantly different from that in the blank group(P<0.05);the absorbance of peritoneal fluid in the high-dose and aspirin groups was significantly different from that in the blank group(P<0.05).The inhibitory effect of Difutongru on chronic inflammation was studied by granuloma test in mice induced by cotton ball.There were significant differences between the blank group and all the Difutongru treated group,and aspirin groups(P<0.05),respectively.In conclusion,high-dose Difutongru showed an obvious inhibitory effect on acute inflammation and chronic inflammation,which is equivalent to the recommended dose of aspirin.Antibacterial test in vitro of Difutongru on common pathogenic of bovine mastitis showed that Difutongru showed a good antibacterial effect on Staphylococcus aureus and CNS,the bacteria is even extremely sensitive to 0.25 g/m L Difutongru.It is a certain antibacterial effect on Escherichia coli,Streptococcus agalactiae,and Streptococcus dysgalactiae.3.Fifty dairy cows with clinical mastitis were randomly divided into 4 Difutongru groups and an antibiotic control group.Four groups,250 m L,500 m L,750 m L,1000 m L Difutongru-treated group,taken Difutongru once a day for 3 to 6 days and antibiotic control group taken Procaine Benzylpenicillin+Gentamycin Sulphate+Dexamethasone Sodium Phosphate twice a day for 1 to 6 days.Clinical results showed that the cure rate of the four groups was 60%,90%,80%,90%,respectively,and the cure rate of the antibiotic group was 90%.LMT test of quarter samples before and after treatment revealed LMT test of cured cows changed from strongly positive to slight positive or suspicious.This study determined the production process of Difutongru,pharmacodynamic effect,and clinical efficacy,which provided data support for pilot experiments and new veterinary medicine applications.
