| Lead(Pb)is a toxic heavy metal that can cause serious pollution to the environment.It is widely used in industrial production and its impact becomes more and more serious on the environment and ecosystem.Lead poisoning can cause damage to human and animal organs.At present,there are many studies of the damage to animal organs caused by lead at home and abroad,but the research of lead on poultry kidney damage is relatively lacking.Selenium(Se)is an essential trace element in the body and can effectively reduce the heavy metals toxicity.However,the effect of Se on Pb-induced kidney damage to chickens is unclear.The aim of this study is to investigate alleviative effect of Se against Pb-induced damage by regulating the expression of antioxidant index,inflammatory factors,cytokines and heat shock proteins;the concentrations of Se and Pb;the contents of creatinine(CREA)and uric acid(UA)in chicken kidneys.In the experiment,one hundred and eighty 7-day-old male chickens were randomly divided into four groups with 45 chickens forming a group.They were respectively fed standard commercial diet(0.49 mg/kg Se)and drinking water;sodium selenite(Na2Se O3)-added standard commercial diet(1mg/kg Se)and drinking water;standard commercial diet and lead acetate((CH3COO)2Pb)-added drinking water(350 mg/L Pb);and Na2 Se O3-added standard commercial diet(1 mg/kg Se)and(CH3COO)2Pb-added drinking water(350 mg/L Pb).On the 30,60 and 90 days,15 chickens were selected from each group,then euthanized them and their kidneys were taken out as samples.Detected the antioxidant index(the activity of T-AOC,GSH-Px,GSH-ST,SOD,CAT and the concentration of GSH,MDA,H2O2);the m RNA expression of inflammatory factors(NF-κB,TNF-α,COX-2 and PTGEs),cytokines(IL-2,IL-4 IL-6,IL-12,IL-12β,IL-17 and IFN-γ)and heat shock proteins(HSP27,HSP40,HSP60,HSP70 and HSP90);the protein expression of NF-κB,COX-2,PTGEs,HSP60,HSP70 and HSP90;Se and Pb concentration;CREA and UA content;histological and ultrastructural changes.The results showed that:(1)Excessive Pb decreased T-AOC,GPx,GST,SOD,CAT activities and GSH content,increased MDA and H2O2 contents in chicken kidneys.(2)Excessive Pb induced NF-κB,TNF-α,COX-2,PTGEs,IL-4,IL-6,IL-12β,and IL-17 mRNA and NF-κB,COX-2 and PTGEs protein expression in chicken kidneys.Excessive Pb inhibited IL-2 and IFN-γ mRNA expression in chicken kidneys.(3)Excessive Pb increased HSP27,HSP40,HSP60,HSP70 and HSP90 mRNA and HSP60,HSP70 and HSP90 protein expression in chicken kidneys.(4)Excessive Pb decreased Se concentration and increased Pb concentration.Excessive Pb damaged the kidney tissue through the functional and morphological aspects.In function,excess Pb increased CREA and UA contents.In morphology,found kidney glomerular fibrosis,renal cysts margin blurred,renal tubular epithelial cell vacuolization,necrosis and other histological changes;lead to chicken kidney mitochondrial swelling,mitochondrial rupture,Deformation and other ultrastructural changes.(5)Selenium relieved the changes in the above indicators of chicken kidney caused by Pb poisoning.The results indicated that excessive Pb can cause oxidative stress,inflammatory response and immune response in chicken kidney.Excessive Pb induced the kidney toxicity in chickens,and Se had a alleviative effect on Pb-induced kidney toxicity in chickens. |
