Lead,one of the highly toxic heavy metals,in nature is a kind of widely used common pollutants.The harm of the lead, small amounts of lead can cause the body’s immune,cardiovascular,reproductive system,and damage to the brain,liver,etc. Selenium,are the essential nutrients,can reduce heavy metals toxicity.At present,the most studies of lead focused on the influence on children’s nervous system,learning,cognitive,and the study of poultry liver function is relatively lacking. So,our experiment divided one hundred eighty Hyline 7-day-old male chickens into four groups: control group,the selenium group,lead and selenium+lead group. Fed commodity diet in the control group,fed commodity diet added selenium in the selenium group,lead group fed commodity diet and drinking tap water,selenium+lead group fed with selenium commodity diet and leaded to drink tap water. In the experiment of 30 days,60 days and 90 days from each group randomly selected 15 samples to euthanasia. Experiment through detected the chicken liver tissue pathological change,the change of the antioxidant index(The activity of SOD,T-AOC,GSH-Px,GSH-S,ATP and the concentration of MDA,GSH,H2O2),the change of the concentration of NO and i NOS activity,and inflammation of the related factors and heat shock protein(NF-κB,TNF-α,i NOS,COX-2,PTGEs,HSP27,HSP40,HSP60,HSP70 and HSP90) m RNA expression,the following results are obtained:(1) the lead poisoning raised chicken liver MDA content,reduced the activity of SOD,T-AOC,GSH-Px,GSH-S,ATP and the GSH,H2O2 conent,selenium relieved lead poisoning caused the change of the antioxidant index;(2) the lead poisoning raised chicken liver i NOS activity and NO content,NF-κB,TNF-α,iNOS,COX-2 and PTGEs mRNA level induced the inflammatory responses,selenium relieved the inflammatory responses induced by lead poisoning;(3) lead poisoning raised the HSP27,HSP40,HSP60,HSP70 and HSP90 mRNA level,selenium relieved lead poisoning caused the increasing of heat shock protein mRNA level;(4) lead poisoning led to the chicken liver damage; selenium can alleviate lead caused the injury.Our research results showed that the lead poisoning caused inflammation responses and oxidative damage,made the HSPs m RNA expression increasing. And selenium relieved the inflammation responses and oxidative damage and the increasing of HSPs m RNA expression induced by lead. |
