Lead(Pb) is toxic heavy metal pollutants widely exists in the environment, as its pollution aggravate gradually. It has affected the environment ecosystem, especially on the influence of wild birds is getting heavier and heavier. Selenium(Se) is necessary for the body’s nutritional trace elements, in the animal play an irreplaceable role in physiological processes. It can alleviate the heavy metal poisoning caused by animal. The aim of this study was to investigate lead-induced toxicity in chicken immune organs and ameliorative effect of selenium. In the present study, 7 days old male chickens were randomly divided into four groups and were provided standard commercial diet and drinking water, Na2 Se O3-added standard commercial diet and drinking water, standard commercial diet and(CH3COO)2Pb-added drinking water, and Na2SeO3-added standard commercial diet and(CH3COO)2Pb-added drinking water, respectively, for 30, 60, and 90 days. Tests were performed on ultrastructure, microscopy, T-AOC, SOD, CAT, GPx, and GST activities, GSH, MDA, and H2O2 contents, and IL-2, IL-4, IL-6, IL-12β, IL-17, IFN-γ,HSP27, HSP40, HSP60, HSP70 and HSP90 messenger RNA expression in chicken thymus, spleens and bursa of fabricius. The results showed that:(1) Excess Pb decreased T-AOC, SOD CAT GPx and GST activities, decreased GSH content, increased MDA and H2O2 contents in chicken immune organs. Pb caused oxidative stress in chicken immune organs.(2) Excess Pb decreased IL-2 and IFN-γ mRNA expression, increased IL-4, IL-6, IL-12β and IL-17 mRNA expression in chicken immune organs. Pb induced immune damage in chicken immune organs.(3) Excess Pb increased HSP27, HSP40, HSP60, HSP70 and HSP90 mRNA expression in chicken immune organs. HSPs protected the chicken immune organs. HSPs can be used as excellent molecular biomarkers in toxicology.(4) Alleviation of excess Pb decreased T-AOC, SOD CAT GPx and GST activities, decreased GSH content, increased MDA and H2O2 contents, decreased IL-2 and IFN-γ m RNA expression, increased IL-4, IL-6, IL-12β and IL-17 mRNA expression, increased HSP27, HSP40, HSP60, HSP70 and HSP90 mRNA expression by Se in chicken immune organs. Alleviation of toxic effect of Pb poisoning by Se on chicken immune organs.. |
