Wheat is one of the three major cereals and the main food crops in our country.And the production of wheat is closely related to economic development,social stability and national security,Though the wheat production in our country is rising recent years,the quality of the wheat is not optimized,and its quality characteristics need further optimizing.So we should make use of high-technique to improve or create new germplasm resources,improve the conversion rate and develop agricultural economy.It's becoming more necessary to adapt to the modern agricultural supply side structure reform and improve people's living standards.In this study,we injected N+ into the seeds of the recipient Yumai 49 by ion beam mediated method,and then soaked the seeds in the whole genomic DNA of the donor oat and hexaploid triticalus respectively.After many years' breeding,we can get a number of very valuable variants.In this study,we chose a batch of special variation materials as the object of study,compared the protein content of the variation materials to the control,parent materials,and then analyzed their agronomic and quality traits,which can help us to screen strains with good resistance to natural disasters and high protein content,and proved that the ion beam mediated transformation technique could be applied to plant genetic breeding and provided new thinking methods to genetic workers.Then we observed and analyzed the variants on drought resistance and finally we carried out SRAP polymorphism analysis of variants and studied the difference between variations.This study consisted of three parts,which were analysise on agronomic quality traits,storage protein content,seedling drought resistance and SRAP Polymorphism of the variants,Through three parts of study,a comprehensive analysis for wheat variations at different levels can help to lay the foundation for further study on the mechanism of variation.1.Analysis of storage protein and agronomic traits of mutanted wheat by ion-beamWe analyzed some agronomic traits of the variations in heading stage and the quality of the seeds in the harvesting period,and found that there were many obvious differences on plant height,panicle length,maize length,chlorophyll content and protein content.After analyzing,we found some variation materials had a significant increase trend in spike length and thousand grain weight,and protein content showed different trends,but wet gluten content in all the variations was higher than that of the compare materials.This indicates that the ion beam can help to produce abundant variations in this batch of wheat variants.2.Analysis of drought resistance of wheat varieties at seedling stage under different drought stress conditionsThe drought resistance of nine materials was analysied by simulating the drought stress conditions with PEG6000.The results showed that the leaf chlorophyll content of the leaves of the variation material changed significantly under different stress conditions,and material E109 of CK group increased significantly.The trend of net photosynthetic rate changed different contains three types,and the stomatal size of the variation material is different under different stress conditions.Overall,the length and width of stomatal have different reduction,and the effect of drought stress on the length is larger than that of the width influences.3.Analysis of SRAP Polymorphism of variantion materialsWe extracted the DNA of wheat variations and screened out the suitable SRAP primer combinations,and then analyzed polymorphism of wheat variants by polyacrylamide gel electrophoresis.It was found that 20 pairs of SRAP primers were screened out in 128 pairs of primer combinations,and 833 polymorphic bands were generated.There were 138.9 polymorphic bands for each variation material on average,and six variation materials with a high polymorphism ratio of 55% or more,indicating that these mutated materials are rich in SRAP polymorphism.The number of specific bands produced by each material varied from 9 to 22,and expansion of material-specific bands successful in the variations,all that lays the basis of further study of variation mechanism.
