Firstly, we reviewed the situation of wheat production and development trend, as well as the principle and application of gene transfer mediated by low energy ion beam. Secondly, we introduced a non-reducing disaccharide--trehalose, which is widespread in nature, and its research advancement on helping plants survive better during enviromental stress. Then, based on trehalose-6-phosphate synthase gene, we did relevant research in order to improve the drought tolerance of wheat.We constructed the plant expression vector with trehalose-6-phosphate synthase gene (tpsl gene) under control of pCAMBIA1303 and testified it by enzymolysis and PCR experiments. After the suitable culture conditions for induction and growth of callus and the optimal selection scheme were established, we finished genetic transformation of wheat mediated by low energy ion beam. After the course of resistant calli selection, we obtained transgenetic plants. And there were ten plants which showed positive in the PCR experiments. When we used different doses and temporal hours of dipping in DNA solution, we got different transformation efficiency. And, it reached 1.20% which was the highest in our experiments. In addtion, in the drought stress experiments, an increase ability of transformed plants in drought tolerance was noted.
