The Rhipicephalus turanicus was named in 1940 by Pomerantzev,the study found that the tick was the medium of Rickettsia,Anaplasmosis,Lyme disease,Tick borne encephalitis virus.The pathogens had brought great damage to the healthy development of animal husbandry.In order to identify the Rhipicephalus turanicus and detect partial tick-borne pathogens in southern Xinjiang.Ticks were collected from 31 sampling sites in southern Xinjiang,The ticks collected were identified by traditional morphological taxonomy.The newly identified ticks were selected as the study subjects,and the genes of ticks 12 S rDNA and 16 S rDNA gene amplification were further selected.Fragments were finally identified by traditional morphological classification and modern molecular biology methods.we used PCR technology,to identify the pathogens Rickettsia of ompA ompB 16 s rRNA g1 tA and scal gene,Anaplasmosis phagocytophilum msp4 gene,A.Ovis 16 S rDNA gene,Babesiosis 18 S rDNA gene,Borrelia miyamotoi 16 S rDNA gene,brucellosis 16 S rDNA gene.The positive PCR products were sequenced,and the results were compared with the NCBI/BLAST platform,Pathogens genotype were determined by homology comparison and phylogenetic analysis.The result shoeed In this study,a total of 1144 suspected Rhipicephalus ticks were Confirmed with R.turanicus,the identification found that there were great differences between the R.turanicus morphology,Most of R..turanicus had the tail.According to detect tick-borne disease,We found that The total positive rate of Rickettsia was 15.3%,The positive rate of OmpA,gltA,sca1,OmpB gene were 16.1%(15/93),12.9%(12/93),16.1%(15/93),16.1%(15/93),MEGA5.0 software was used to construct phylogenetic tree,and the results showed that there were three kinds of Rickettsia Candidatus Rickettsia barbariae,Rickettsia massiliae,Rickettsia africae,System evolutionary relationships display Candidatus Rickettsia barbariae constituted a separate branch,Neither of the spotted fever rickettsia SFG nor the typhus group TG.;The total positive rate of Anaplasmosis was 15.3%,The positive rate of A.Ovis and A.Phagocytophilum were9.68%(9/93)and 19.35%(18/93);The positive rate of Babesiosis and Borrelia miyamotoi were 9.68%(9/93)and 3.32%(3/93);Rickettsia africae and Borrelia miyamotoi were firstly identified Nucleic acid positive from the southern Xinjiang R.Turanicus,and it was the firstly reported in southern Xinjiang,No positive samples was detected in Brucellosis.In this study,We combined with the traditional species identification and molecular biology methods to identify accurate species in southern Xinjiang,and we detected Rickettsia,Anaplasmosis,Babesiosis,Borrelia miyamotoi and Brucellosis nucleic acid PCR,Study on the presence and prevalence of these pathogens in ticks by molecular biology and molecular epidemiology,To provide a basis for scientific prevention of ticks.
