In this study,nine cultivars were used as experimental materials,and the field cultivation and horticultural traits were observed in Fuzhou and.the physiologica indexes were measured.The DNA identification was carried out by DNA barcoding.The main findings are as follows:1.The introduction of nine Narcissus pseudonarcissus could adapt to the climate and environment in Fuzhou,after the introduction of the first year in addition to Las Vegas,the normal flowering,flowering from mid to late February to mid to late April,which early flower varieties Dutch Master And the Ice Follies were about 35 days earlier than the late flower varieties Tahiti,Bridal Crown and Salome.Compared with the local Chinese narcissus,budding period of about 35 d,flowering about 30 d later.2.The leaf hatch size,flower size,flower type and scape of nine cultivars were observed in 11 horticultural cultivars,and the results were analyzed by principal component analysis.The results showed that:(1)In these nine varieties.the leaf size,petal size,flower size,inflorescence type and several different characteristics of scape were significant,in which Tahiti’s scape was the highest;Dutch master’s petal aspect ratio was the largest;Bridal Crown’s inflorescence is multicolored and others are single-edged;Tahiti,Bridal Crown,Dick Wilden and Replete petals are reed;(2)The principal component analysis method obtained three principal components,the contribution rate of the first principal component variance was 59.7%,which mainly reflected petals traits.The contribution rate of the second principal component variance was 20.1%,which mainly reflected the corolla traits And the proportion of the third principal component was small,which mainly reflects the leaf traits.The evaluation model of the natural horticultural characteristics of the daffodils was established by using the variance contribution rate of the three principal components as the coefficients.Ice Follies,Replete,Dutch Master,Tahiti,Pink Charm,Salome,Dick Wilden,Bridal Crown,Las Vegas,among which the most ornamental variety wad Ice Follies.3.The physiological indexes such as chlorophyll and soluble protein were measured in the leaves of eight Narcissus pseudonarcissus varieties in the blooming stage,and the correlation between the results and the ornamental traits was analyzed.The results showed that:(1)There were significant differences in chlorophyll,soluble protein and proline content,superoxide dismutase and peroxidase activity among different cultivars(P<0.05).(2)Correlation analysis showed that there was a significant negative correlation between leaf width and leaf number,flower number and leaf length and flower length and leaf width(P<0.05);leaf width and flower diameter,leaf width(P<0.05).POD activity and leaf number,POD activity and leaf shape index(P<0.05)were significantly higher than those of petal length,petal width and flower diameter,petal width,petal length and secondary crown diameter and height and diameter of crown(P<0.05).There was a significant positive correlation between POD activity and height of scape and Pro content and height of crown.There was a significant negative correlation between POD activity and secondary height,POD activity and crown diameter and POD activity and height/diameter of crown(P<0.01).There was a significant positive correlation between leaf shape index and leaf number,flower diameter and petal length and height of vice crown and height/diameter of vice crown(P<0.01).4.The DNA identification of 9 Narcissus pseudonarcissus cultivars and Narcissus pseudonarcissus were studied by DNA barcoding.The results showed that:(1)the tree construction method was used to analysis the results,the chloroplast gene fragment has the ability to accurately identify the foreign narcissus,and can recognize the genetic relationship among the foreign narcissus species,but can not distinguish between Chinese narcissus and foreign narcissus;the gene sequence of the nuclear gene fragment can accurately identify the Chinese narcissus and the foreign narcissus,but it can not accurately identify the genetic relationship in the identification of the genetic relationship between the foreign narcissus species;The four gene fragments have a strong ability to identify,can accurately identify the Chinese narcissus and foreign narcissus,but also to identify the genetic relationship between foreign narcissus species,and can identify the Chinese narcissus and foreign daffodils have a certain genetic relationship;(2)From the point of view of distance analysis,the combination of ITS + matK,ITS + trnH-psbA,ITS+ rbcL and ITS + matK + trnH-psbA + rbcL is suitable for the identification of Narcissus species. |
