Cucumber is belonged to cucurbitaceous,it is one of the most important vegetable crops in the world. Along with the improvement living standards and change of diet,people begin to pay attention to the function of food.Because Fruit Cucumber rich of alcohol acid, cucumber enzyme and other active substances, and a lot of vitamin E.It's quickly became the best-selling products on the market,To develop different function food market is optimistic. Momordica grosvenori with treatment of hypertension,hyperlipidemia,obesity and other effects,it also rich of glycoside and has hypoglycemic effect,it can help auxiliary treatment and obesity.Fruit cucumber and Momordica grosvenori all belong to the cucurbitaceae,so it is possible to let siraitia grosvenorii genomic DNA integrated into fruit cucumber,to obtain containing glycosides fruit type cucumber. In this study,the total DNA as the donor of siraitia grosvenorii,with big fruit type cucumber fruit and small fruit type cucumber as receptor. Momordica grosvenori DNA was transformed into fruit cucumber by 3 introduction mothods which are ovary injection* pollen tube pathwayand ovary injection combined with pollen tube pathway. Comparing of 3 methods of transformation descendants percentage of fruits,the results as below.(1) Use ovary injection?pollen tube pathwayand ovary?injection combined with pollen tube pathway get different concentrations of Momordica grosvenori DNA into two kinds of fruit cucumber. Comparing of 3 methods of transformation the effect of the offspring fruit, the fruit type cucumber pollen tubepathway into the average fruit set rate was 59.9%,small fruit type 55.1%, the DNA concentration is 187.316?g/ml, big fruit type cucumber average fruit set up to 71.4%, the concentration of DNA for 234.145?g/ml, average fruit type cucumber fruit reached 68.8% The results showed that the pollen tube pathway method was superior to ovary injection and ovary injection combined with pollen tube pathway method, which could provide a useful basis for the genetic transformation of plants.(2) By comparing the plants and fruit of fruit cucumbers D1 generation with two controls, the two fruit cucumber Dl generation development were slower than the control groups. In the small fruit cucumber D1 generation, mutant plants was found obviously. Provide materials for further screening glucoside containing high amount of varieties.(3) Through the pollen tube pathway Siraitia DNA into Cucumber, detect related physiological and biochemical indexes of offspring.The results showed that the contents of glucose in the descendants of two fruit-cucumbers reduced 0.2% and 0.22% respectively.The contents of protein increased 0.381% and 0.375% respectively.(4)The photosynthetic characteristics of the leaves of the donor, recipient and the offspring were determined, and the net photosynthetic rate, stomatal conductance, transpiration rate and intercellular CO2 concentration were determined by the light and the donor.
