Momordica grosvenori is a traditional medicinal and edible plant in our country. Mogroside, a kind of natural sweeteners from Momordica grosvenori, is suitable for the diabetic patients by its high sweetness, low carorie, good taste and without any side effects. Mogroside become the raw material of many food and nutriment, more and more widely used. In the early 1980s, a kind of mogroside with high sweetness extracted from Momordica grosvenori. named Mogroside V. The research shows that MogrosideⅤis a excellent natural sweetener. Its suger content just about 1%, but the sweetness equivalent to 300 times of sucrose. MogrosideⅤis the main components of Mogroside, so the content of MogrosideⅤplays the decisive part in product's quality of glycoside from Momordica grosvenori.At present, Momordica grosvenori sweeteners at the market are mainly extrat of Mogroside, the product of MogrosideⅤis expensive and rarely sold, that because of immature technologies of extraction and purification of high purity MogrosideⅤfrom Momordica grosvenori. So this paper focus on the extraction and purification of high purity Mogroside V.The paper research on dry fruit of Momordica grosvenori, establish the HPLC to detect the content of Mogroside V in different parts of Momordica grosvenori, make further study on optimum extraction technology, confirm the parameters, explore chromatography purification technology of macroporous resin and silica gel column to prepare for practical production. The results are as follows:1. The HPLC detects the contents of MogrosideⅤby quantitative analysis. The conditions of determing liquid chromatogram are as follows:chromatogram column Hypersil ODS (250 mm×4.6 mm,5μm), temperature at 35℃, the wavelength of detector at 203 nm, sample size 20μL,rate at 0.8 mL/min, acetonitrile:water(23:77v/v) as mobile phase.2. The content of Mogroside V in different parts of Momordica grosvenori were measured by HPLC. The results showed that Mogroside V was existed in different parts. The content of Mogroside V in Momordica grosvenori dry fruit was 0.22%, which was the highes, while in root was 0.02%, which was the lowest.3. Explore the optimum extraction technology. Mogroside V was extracted from Momordica grosvenori by ethanol heat reflux extraction, through oneway and orthogonal experiment. The optimum parameter of extraction had been determined: the concentration of ethanol is 50%, the multiple of ethanol to material is 1:20, extraction time is 120 min, extraction temperature is 70℃, by two extractions, the rate of extraction was 72%. It was also showed that the main factor influencing extraction efficiency was ethanol concentration, while the factor was extraction time.4. The purifying technology of Mogroside V. The study compared the adsorption properties of six macroporous resin, including D101,D201,HPD400,HPD100,NKA-1,AB-8, and the resin AB-8 was determined as the best sorbent. The optimum purification condition obtained was that the Mogroside crude extract which concentration was 0.4 mg/mL and flow rate on desorption was 2 BV/h was successively eluted with water,25% ethanol and 60% ethanol at pH 9.0, and then 60% ethanol eluate was collected. The purity and the yield of Mogroside V in 60% ethanol eluate was 52.2% and 61% determined by HPLC. The high purity Mogroside V which purity was up to 89.6% was obtained by silica gel column layer chromatography, which yield was 62.3%.
