| Frogeye leaf spot of soybean occurs worldwide and is one of major soybean disease in northeast of China, harming soybean seriously.The most effective and economical way is planting resistance cultivars.It was found the Cercospora sojina Hara races a lot, in recent years, the development trend of race 12 faster, and the study about race 12 is little.In this experiment, 320 kinds of common soybean varieties were infected by the Cercospora sojina race 12, and carried out the resistance identification. Genomic scanning,population structure and linkage disequilibrium analysis of soybean materials using 300 pairs of SSR primers, asoybean resistance association analysis run with software STRUCTURE and GLM procedure from software TASSEL. The results were as follows.First, resistance of 320 materials were identified, the results were divided into 5 grades :there are 23 varieties were high resistance, accounting for 7.19% of the total number of materials; 53 varieties were resistance to disease, accounting for 16.56% of the total number of materials; 178 varieties were resistance, accounting for 55.62% of the total number of materials; 56 varieties were susceptible, accounting for 17.5% of the total number of materials;10 varieties were susceptible, accounting for 3.13% of the total number of materials.Second, population structure analysis of 320 kinds of soybean varieties by STRUCTURE,and divided into 3 sub-populations,the number of materials were 163, 69, 88.Third, 300 SSR markers generated a total of 1175 alleles, average number of effective alleles per locus is 3.92, PIC ranged from 0.015 to 0.836 on an average of 0.510.Forth, using the general linear model procedure of Tassel 3.0 software, the correlation regression analysis was carried out on the resistance phenotype data of Cercospora sojina Hara and 300 SSR molecular markers.The results showed that the resistance of soybean to Cercospora sojina race 12 was 69, and the Satt365 on C2 linkage group was the highest,reaching 3.22%. 12 loci showed extremely significant correlation and 47 loci showed significant correlation.Fifth, Satt164-6 and Satt365-2 is the max of positive phenotypic effect(+9.09)and the max of negative phenotypic effect(-30.68), respectively.Among the related loci, positive allele effect of Satt156 is the highest(+25.13), and negative allele effect of Satt365 is the highest(-35.97).The last, 12 loci were found in this experiment: Satt534 on the B2 linkage group;Satt399、Satt164 on the C1 linkage group; Satt422、Satt365、Satt202 on the C2 linkage group; Satt580 on the D1 a linkage group; Satt335、Satt510 on the F linkage group; Satt052 on the H linkage group; Satt156 on the L linkage group; Satt590 on the M linkage group. |
