Physiologic Race Differentiation And Genetic Diversity Of Cercospora Sojina From Soybean In Heilongjiang Province

Soybean frogeye spot disease is a plant fungus disease which is infected by Cercospora sojina Hara. It causes serious hazard in production. The main reason of the loss of resistance to Cercospora sojina is the variety of Cercospora sojina. So, inspecting the change law of Cercospora sojina is the basis of control the disease.337 isolates of soybean frogeye fungus, Cercospora sojini,were collected in Heilongjiang province from 2006 to 2009.We identificated the 337 isolates by using 6 differential soybean varieties, and 121 isolates form 337 isolates were used to analyze the genetic diversity of cercospora sojina.the results were as followed:1.16 physiologic races were identified using 6 differential soybean varieties during 2006-2009. 6 new physiologic races (No.5, No.12, No.13, No.14, No.15 and No.16) were found, apart from reported 10 physiologic races (No.1, No.2,No.3, No.4, No.6, No.7, No.8, No.9,No.10 andNo.11).2.The race No.1 was the highest frequent one with the frequency of 39.17%, and race No.7 was the second highest one with the frequency of 14.84%, and then was race No.10 with the frequency of 7.72%.Others were respectively No.2 (5.34%), No.3 (5.04%), No.6(5.04%), No.4 (4.15%), No.9 (3.86%), No.8 (3.56%), No.ll (2.97%), No12 (2.08%), No.5 (1.78%), No.14 (1.48%),No.13 (1.19%),No.15 (0.89%) and No.16 (0.89%).3.Race No.1 was the dominant one in the main soybean areas of Heilongjiang provinc e, types and compositions of the race were different from place to place.There were No.l, No.2,No.3,No.7,No.9 and No.10 physiologic race in Harbin district; No.l,No.2,No.3,No.4,N o.6,No.7,No.8,No.9,No.10,No.11and No.12 were the mainly races in Qiqihaer district; Mudan jiang district had No.1,No.2,No.3,No.6,No.7,No.8,No.10,No.11,No.15 and No.16 physiologi c race;No.1,No.2,No.3,No.4,No5,No.6,No.7,No.8,No.9,No.11,No.12,No.14 and No.15 physiolo gic races were in Jiamusi district;While physiologic race types in other districts were respe ctively that Suihua district (No.1,No.2,No.3,No.4,No.5,No.6,No.7,No.8,No.9,No.10,No.11,No.1 2 and No.14),Heihe district(No.1,No.2,No.3,No.4,No.7,No.9,No.10,No.12 and No.13),Yichuan district(No.1,No.2,No.4,No.8 and No.12),Jixi district(No 1,No 6,No 7,No 10 and No 11),Qit aihe district (No.1,No.2,No.6 and No.7),Hegang district(No 1,No 2,No 7,No 8 andNo 10), Daqing district(No.1,No.4,No.7 and No.10),Shuangyashan district(No.1,No.6 andNo.7),Daxin ganling district(No.1,No.7 and No.12).4.10 higher Polymorphism and stable pairs of primers were selected from 100 pairs of AFLP primers by using DNA of 8 typical isolates of cercospora sojina.148 different framents were obtained after AFLP analysis of 121 cercospora sojina isolates from Heilongjiang province were done.lll different cell types of the representative AFLP fingerprint were aquired by polyacrylamide gel electrophoresis.0.81 was as the genetic distance boundary,121 strains were divided into 6 genetic lingeages by Cluster analysis using UPGA software. L03, L05, L06 were the dominant Genetic lingeages in Heilongjiang province. Geographical distribution of L02 was relatively narrow, and it was a rarely genetic lingeages of cersospora sojina in Heilongjiang province.5.Cercospora sojina in Heilongjiang province showed rich genetic diversity, there were obviously different in the 6 genetic lingeages. L01 distributed in middle area of Heilongjiang province,including Harbin,Suihua districts; L02 was in the Southeast of Heilongjiang province, including Mudanjiang,Jixi and Qitahe districts; L03 was in the east of Heilongjiang province, including Jamusi,Shuangyashan,Hegang and Yichun districts, and it rarely existed in the Southeast & the east of Heilongjiang province; L04 was in the east,north and middle of Heilongjiang province; L05 was mainly in the west and the middle of Heilongjiang province,including Qiqihaer,Daqing disitrict; L06 existed mainly in the North and the middle of Heilongjiang province.
