China is a country with large amount of pigeon production,according to the statistics,about 40 million pairs of pigeon are raised and the annual output of squab is about 600 million,which produced production valve of 10 billion annually.At present,meat-type pigeon breeding is lagging behind other poultry,and lack of national pigeon breed.The key of enhance pigeon industry is strengthen the pigeon breeding.L-FABP gene is considered to be important in the lipid metabolism for the gene participating in uptake,transport,mitochondrial oxidation and esterification of the fatty acid.In the present study,the genetic variation of L-FABP is identified and the genetic effects of the variation were investigated,which is helpful in molecular breeding for pigeon.28-day-old White King(n=100)was randomly selected.According to the NCBI(Gene ID:102086798),seven pairs PCR primers were designed.PCR products covered 5’-region(-1162 ~ 0)and 4 exons promoter and part of the introns region in L-FABP.By direct sequencing of PCR products,five SNPs were identified: 100 G > C in exon 1,and the G,C allelic frequencies is 0.38 and 0.62.1237 T > C is in exon 2,and the T,C allelic frequencies is 0.67 and is 0.33.1327 T > C located on exon 2,and the T,C allelic frequencies is 0.64 and 0.36,and 100 G > C,1237 T > C,and 1327 T > C mutations are synonymous.3838 C > T located in intron 3,and the C,T allelic frequencies is 0.58 and 0.42.3852 G > T located in intron 3,the G,T allelic frequencies is 0.58 and 0.42.Genetic characteristics analysis showed that each SNP site was moderate polymorphic(0.25 <PIC <0.50),and was in the Hardy-Weinberg equilibrium.For each SNP,the association between genotype and squab growth traits and carcass traits: in 100 G > C,abdominal fat rate of genotype GG(1.2%)and GC(1.3%)was significantly higher than(P <0.05)CC(0.7%);in 1327 T > C,breast muscle rate of genotype TT(17.7%)and TC(16.6%)was significantly lower than(P <0.05)CC(19.3%),and leg muscle rate of TT(9.8%)and TC(9.2%)was significantly lower than(P <0.05)CC(10.8%).In conclusion,5 SNPs were identified in L-FABP in White King,in which 100 G > C associated with abdominal fat rate,and 1327 T > C associated with breast muscle rate and leg muscle rate in squab.
