Wheat variety,ZHENG 9023 which has high levels of gluten were radiated by 60Co-? ray with the dose of 200 Gy.The genetic variation analysis of No.of degradation of spikelet,plant height,No.of spike,spike length,No.of spikelet,TKW,grain yield etal.7 agronomic traits and unit weight,stabilization,formation,flour yield,amylase,wet gluten content,protein content,hardness,sedimentation,GMP content,GMP/Pr,PPO activity,LOX activity etal.13 quality traits of 918 accessions in M2 population and 166 lines in M3 population were carried out,and the mutants were identified by sodium dodecyl sulphate?SDS-PAGE?.Identification of M3 population by SSR.The result showed as follows:1.Genetic variation analysis of agronomic traits showed: In M2 population,panicles,No.degradation of spikelet,grain yield and content of amylase have wealth of genetic variation ability and good selection effect.TKW,stabilization,formation,sedimentation,GMP content,GMP/Pr and PPO activity have moderate variation abilities;plant height,spike length,No.of spikelet,unit weight,flour yield,wet gluten content,protein content and hardness have low coefficient of variation and stable genetic capability.The results showed that depending on the criterion of over meaną1.96 SD,there are selecting variation plant of 7 agronomic traits.Plant yields only have positive variation,no negative variation.The highest negative mutant rate of TKW is 4.9%.In M3 population,plant height has 1line in positive variation and 3 lines in negative variation;spike length has 2 lines in positive variation and 5 lines in negative variation;No.of spike has 4 lines in positive variation and 1 line in negative variation;No.degradation of spikelet has 3 lines in positive variation and 5 lines in negative variation.2.The population mean of M2 population of sabilization,formation,amylase,wet gluten content,hardness,sedimentation and PPO activity are lower than parents,and the population mean of flour yield,protein content,GMP content and GMP/Pr are higher than parents.The results of correlation analysis indicated that plant height is positive correlation with most quality traits;plant height has not significant correlation with GMP/Pr and PPO activity;protein content has positive correlation with wet gluten content;PPO activity has positive correlation with unit weight and stabilization.The population mean of M3 population of protein content,wet gluten content,stabilization,formation and LOX activity are higher than parents;Unit weight,hardness,sedimentation,amylase,GMP content,GMP/Pr and PPO activity are lower than parents.The results of correlation analysis indicated that amylase is negative correlation with LOX activity,GMP content is notable positive correlation with GMP/Pr,GMP content is notable negative correlation with PPO activity,GMP/Pr is notable negative correlation with PPO activity,PPO activity is correlation but unnotable with LOX activity.3.High-molecular-weight glutenin subunit?HMW-GS?composition have been examined by using sodium dodecyl sulfate polyacrylamide gel electrophoresis system.M2 population and M3 population have 6 HMW-GS composition variation plants from?N/7+8/2+12?to?1/7+8/2+12?,respectively,quality scores increased 2 points.By phenotypic identification and HMW protein subunits identified,considering the yield and quality factors,variation plant 225 is better than the other variation plants.There are 13 variation lines of high gluten in M3 generation population.Considering the yield and quality factors,variation line 161 and 164 is better than the other variation lines.4.According to SSR identification indicated,detected mutation sites in wheat chromosome A,B,D of M3 generation population.There are 12 minor genetic markers of 918 accessions in M3 population.In M3 progenies of Zheng9023,mutation occurred in genome 3B?2A?2D?3A?4A?1B?7B?5B.There were 94 SSR locus which mutated?56.6%?in genome A,104 SSR locus which mutated?62.7%?in genome B,12 SSR locus which mutated?7.2%?in genome D.It shows that the genome mutant frequency is different in M3 population of Zheng9023,it's B> A >D.
