Rosa roxburghii Tratt is a species of medicinal and edible wild plants,which was published as one of the earliest new varieties of food resources by Health Planning Commission,with efficacy of convergence and antidiarrheal,which could promote digestion and strengthen spleen,mainly used for the treatment of indigestion feed abdominal distension,diarrhea and other diseases.Recently,research showed that R.roxburghii Tratt displayed variety biological activities,including antioxidant,anticancer,cancer prevention,reducing the heavy metal load,regulating body immune,sedation,anti-aging,anti-atherosclerosis activities.Therefore,there comes up a variety of products,among them were extract such as juice for the most of market sales.As a new food resource,with medicinal and extract,there was no standard extraction preparation and quality evaluation methods.In view of this,this paper developed extraction process of standard extract on the basis of determination method of total triterpenes,total flavonoids and vitamin C,and evaluated the standard extract quality by antioxidant activity in vitro.The main research contents and results were as follows:1.The content of vitamin C in fresh fruit of Rosa roxburghii Tratt was determined by UV-visible spectrometry method and 2,6-dichloro-indophenol titration.Results of the two methods show great differences for determining Vc.The former was 2.83 mg/mL,the latter was 15.93 mg/mL,and both were in average content.The content of total flavonoids in extract which was determined by UV method was 24.73%(ethanol extract R-C),57.90%(total triterpenes R-S).The content of total flavonoids in extract was very low and hardly detected by UV method.2.The effect of ethanol concentration,solid-liquid ratio,extraction time and times on extraction of total triterpenes in R.roxburghii Tratt was studied by single factor experiment and orthogonal test,and the content of total triterpenes was regarded as the detection index.Results showed that the optimum process conditions of extraction were as follow: reflux extracted 1.5 h,3 times with 15-fold 75% ethanol.3.This paper studied antioxidant activity in vitro by Uv-vis spectrophotometry and WST-1 method,R.roxburghii Tratt and its extracts scavenging DPPH,hydroxyl,superoxide radical were evaluated,as well as determination of SOD in R.roxburghii Tratt.The results showed different parts of extracts exhibited strong scavenging effects on DPPH radical,concentrations vary from 0.02 to 0.2 mg/mL,the antioxidant capacity was R-C> R-S> Vc> water-soluble parts(R-L),respectively,between the mass concentration of 0.4-0.8 mg/mL was R-S> Vc> R-C> R-L.The experiment results showed that extracts of R.roxburghii Tratt exhibited weaker hydroxyl radical-scavenging than control ascorbic acid,scavenging capacity was Vc> R-S> R-C> R-L.Between the mass concentration of 1.0-3.0 mg/mL,the capacity scavenging superoxide radical was the same with hydroxyl radical.The results of WST-1 method showed that tissue sample solution of R.roxburghii Tratt on SOD inhibition rate increases with the dilution ratio,the inhibition rate reached the maximam 97.24%,then began to decrease,the activity of SOD increased with the dilution ratio,reached to 1547.6U/g when 10 fold dilution.In summary,the detection method is simple,accurate and feasible,can be used for quality control of the corresponding components in R.roxburghii Tratt.The research process is scientific,reasonable,can be used for industrial produution.The evaluation quality is objective,safety,to provide reference for further rational development of R.roxburghii Tratt.
